姜黄素基于下调miR210与TLR4/NF-κB信号通路表达对人前列腺癌PC3细胞株凋亡的影响 被引量:6

Effect of Curcumin on Apoptosis of PC3 Cell Line via Down-Regulating the Expressions of MiR210 and TLR4 /NF-κB Signaling Pathway

摘要 目的:观察姜黄素是否通过下调miR210表达,抑制TLR4/NF-κB信号通路关键因子表达促进人前列腺癌PC3细胞株凋亡。方法:检测人前列腺PC3细胞荷瘤小鼠肿瘤重量;前列腺癌PC3细胞株培养后以姜黄素10、20、40μmol/L处理48 h,CCK-8法检测PC3细胞株细胞增殖情况;AnnexinV-FITC/PI检测细胞凋亡情况;qRT-PCR检测miR210相对表达量;ELISA检测IL-6和TNF-α含量;Westernblot检测Toll样受体4(TLR4)、核转录因子κB(NF-κB)、B细胞淋巴癌-2(Bcl-2)、Bax蛋白表达及Bcl-2/Bax比值。结果:与荷瘤模型对照组比较,姜黄素50、100、200 mg/kg组显著抑制肿瘤重量,显著升高抑瘤率(P<0.01)。与PC3细胞株模型对照组比较,姜黄素10、20、40μmol/L能明显抑制PC3细胞增殖,增加肿瘤细胞凋亡率,降低Bcl-2/Bax比值,下调miR210表达,下调TLR4、NF-κB蛋白表达,降低IL-6和TNF-α含量(P<0.05或P<0.01)。结论:姜黄素能抑制PC3细胞增殖,促进其凋亡,减轻裸鼠鼠移植肿瘤重量,机制可能与下调miR210表达后抑制TLR4信号通路,减少炎症因子表达有关。 Objective: To investigate the effect of curcumin on the apoptosis of PC3 cell lines via regulating the expression of miR210 and TLR4/NF-κB signaling pathway.Methods: The tumor weight was measured in mice transplanted with human prostate PC3 cells. PC3 cell were incubated with curcumin at the doses of 10,20,40 μmol/L for 48 h, the cell proliferation of PC3 cells was detected by CCK-8, the apoptosis of PC3 cells was detected by AnnexinV-FITC/P, the expression of miR210 was detected by qRT-PCR, the expressions of IL-6 and TNF-α were detected by ELISA, the protein expressions of TLR4, NF-κB, Bcl-2, Bax and ratio of Bcl-2/Bax were detected by Western blot.Results: Compared with the model group, the tumor weight was significantly inhibited in 50、100、200 mg/kg curcumin groups, the anti-tumor rate was significantly increased(P<0.01). Compared with PC3 cells control group, the PC3 cell proliferation was significantly inhibited in 10、20、40 μmol/L curcumin groups, the apoptosis rate was increased, the ratio of Bcl-2/Bax was decreased, the expression of miR210 was reduced, the protein expressions of TLR4, NF-κB and the contents of IL-6 and TNF-α were decreased(P<0.05 or P<0.01).Conclusion: Curcumin can inhibit the proliferation of PC3 cells and promote the apoptosis, reduce the weights of transplanted tumor in nude mice. The mechanism may be related to inhibiting TLR4 signaling pathway, down regulating Mir210 expression and reducing the expressions of inflammatory factors.
