摘要
目的通过腹腔注射链脲佐菌素(STZ)及喂养高脂高糖饲料+注射STZ分别建立1、2型糖尿病大鼠模型,观察2种类型糖尿病大鼠视网膜病变情况及血清抗氧化指标、视紫质(RHO)蛋白含量的变化。方法45只SD大鼠随机分为三组:对照组15只,按60 mg/kg腹腔注射柠檬酸缓冲液;模型Ⅰ组15只,按60 mg/kg剂量一次性注射2%STZ柠檬酸缓冲液;模型Ⅱ组15只,高脂高糖饲料喂养30 d后,按45 mg/kg剂量一次性注射2%STZ柠檬酸缓冲液。注射后第1、3、7、14、30、60天检测大鼠空腹血糖,裂隙灯显微镜观察大鼠视网膜病变情况,60 d后采集血清,检测血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)和RHO含量。结果模型Ⅰ组视网膜病变发生率为91.7%,模型Ⅱ组为92.3%;与对照组比较,模型Ⅰ、Ⅱ组大鼠血清SOD、GSH-Px活性均显著降低,MDA含量显著升高,RHO含量显著降低,差异均有统计学意义(P<0.05),且模型Ⅰ、Ⅱ组大鼠SOD、GSH-Px、MDA、RHO比较,差异均无统计学意义(P>0.05)。结论2种方式建立的糖尿病大鼠模型均诱导了较高的视网膜病变发生率,可以作为糖尿病性眼病的研究模型,且该病变的发生与血清抗氧化水平的降低有显著相关性。
Objective To observe the rate of retinopathy and test serum antioxidant index and content of retinal purpurin in two types-diabetes rats mode,we established the model of type I and type II diabetic rats by Streptozotocin(STZ)injection intraperitoneal and feeding high fat and high sugar diet plus STZ injection.Methods Forty-five SD rats were randomly divided into three groups.Control group,15 rats were given one-time injection of citric acid buffer with 60 mg/kg.Model group I,15 Rats were given one-time intraperitoneal injection with 60 mg/kg 2%STZ buffer.Model group II,15 Rats were fed with high fat and high sugar diet for 30 days,then given one-time intraperitoneal injection with 60mg/kg 2%STZ buffer.After injection,fasting blood glucose was measured on 1st,3rd,7th,14th,30th and 60th day.Retinopathy was observed with slit lamp microscope,and SOD,GSH-Px,MDA in serum and rhodopsin in retina were detected.Results The incidence of retinopathy in model groupⅠwas 91.7%,which in groupⅡwas 92.3%.Compared with the control group,the activity of SOD and GSH-Px in the serum of the model groupⅠandⅡdecreased significantly(P<0.05),the content of MDA increased significantly(P<0.05),and content of rhodopsin in retina of both model group were increased significantly(P>0.05).Conclusion Both models of diabetic rats induced a high incidence of retinopathy,which can be used as the research model of diabetic ophthalmopathy,and the incidence of diabetic retinopathy has a significant correlation with the decrease of serum antioxidant level.
