滋膵降糖方对2型糖尿病合并非酒精性脂肪性肝病大鼠胰岛素抵抗的影响 被引量:1

The Influence of Zicui Jiangtang Formula on Insulin Resistance of Rats with Type 2 Diabetes Mellitus Combined with Nonalcoholic Fatty Liver Disease

摘要 目的:观察滋膵降糖方对2型糖尿病(type 2 diabetes mellitus,T2DM)合并非酒精性脂肪性肝病(nonalcoholic fatty liver disease,NAFLD)大鼠胰岛素抵抗的影响,并探讨其作用机制。方法:从54只SD大鼠中随机选择44只大鼠进行造模,另外10只大鼠作为正常组。高脂高糖喂养44只SD大鼠6周后,给予腹腔注射链脲佐菌素40 mg·kg^-1,建立T2DM合并NAFLD大鼠模型。将造模成功的40只大鼠按照随机数字表法分为模型对照组、二甲双胍组、滋脺降糖方组、二甲双胍联合滋膵降糖方组,每组各10只。二甲双胍组给予二甲双胍混悬液150 mg·kg^-1灌胃;滋膵降糖方组给予滋膵降糖方溶液10 mL·kg^-1灌胃;二甲双胍合滋膵降糖方组给药为以上两组的联合用药;正常组、模型对照组分别给予等量生理盐水灌胃,每日1次。测定各组大鼠给药后体质量、空腹血糖(fasting blood glucose,FBG)、空腹胰岛素(fasting insulin,FINS)水平,计算胰岛素抵抗指数(homeostasis model assessment for insulin resistance,HOMA-IR),检测肝脏肝糖原(hepatic glycogen,Glu)、三酰甘油(triacylglycerin,TG)、游离脂肪酸(free fatty acids,FFA)含量,HE染色观察肝脏组织形态学变化,免疫组化法检测肝脏瘦素(leptin,LP)表达。结果:各组大鼠体质量均低于模型组,正常组大鼠体质量低于模型组、滋膵降糖方组、二甲双胍组,差异有统计学意义(P<0.05)。二甲双胍组、滋膵降糖方组、二甲双胍合滋膵降糖方组FBG、HOMA-IR水平高于正常组、低于模型组,二甲双胍组、滋膵降糖方组FINS水平高于正常组,差异均有统计学意义(P<0.05)。二甲双胍组、滋膵降糖方组Glu、TG、FFA高于正常组,差异具有统计学意义(P<0.05),二甲双胍组、滋膵降糖方组、二甲双胍合滋膵降糖方组Glu、TG、FFA水平低于模型组,差异具有统计学意义(P<0.05);二甲双胍合滋膵降糖方组Glu、TG、FFA水平低于二甲双胍组、滋膵降糖方组,差异具有统计� Objective:To observe the influence of Zicui Jiangtang Formula(滋膵降糖方,ZCJTF)on insulin resistance(IR)of rats with type 2 diabetes(T2 DM)mellitus combined with nonalcoholic fatty liver disease(NAFLD),and probe into the mechanism of action.Methods:A total of 44 SD rats were randomly selected from 54 SD rats for modeling,and the other 10 rats were selected into the normal group.The above 44 rats were fed with high fat and high glucose for 6 weeks.An amount of 40 mg·kg^-1 streptozotocin was injected intraperitoneally to establish T2 DM combined with NAFLD rat model.According to the random number table method,40 rats were divided into the model control group,the metformin group,the group of Zicui Jiangtang Formula(滋膵降糖方,ZCJTF),the group of metformin combined with ZCJTF,with 10 rats in each group.The metformin group was treated with 150 mg·kg^-1 of metformin suspension for gavage.The ZCJTF group was treated with 10 mL·kg^-1 of ZCJTF solution for gavage.The group of metformin combined with ZCJTF was treated with the combination of the above two groups.The normal group and the model control group were given 10 mL·kg^-19 ng·L-1 of normal saline by gavage respectively,once a day.Then the body weight,fasting blood glucose(FBG)and fasting insulin(fins)levels were measured.The homeostasis model assessment for insulin resistance(HOMA-IR)was calculated.The contents of hepatic glycogen(Glu),triacylglycerol(TG)and free fatty acids(FFA)were detected.HE staining was used to observe the morphological changes of liver tissue and the expression of leptin(LP)was detected by immunohistochemistry.Results:The body weight of rats in each group was lower than that of the model group,while that of the normal group was lower than that of the model group,the ZCJTF group and the metformin group,and all the differences were statistically significant(P<0.05).The levels of FBG and HOMA-IR of the metformin group,the ZCJTF group,and the group of metformin combined with ZCJTF were higher than those of the normal group but lower
