胃癌组织中NOB1蛋白表达及其与顺铂化疗敏感性的临床关系探讨 被引量:2

摘要 目的探究胃癌组织中酵母基因Noblp人类同源基因(NOB1)蛋白表达及其与顺铂化疗敏感性的临床关系。方法选取2017年5月至2019年6月于衡水市第五人民医院行胃癌根治术的90例胃癌患者作为研究对象,采用免疫组化SP法检测胃癌组织、癌旁组织中NOB1蛋白表达情况,评价患者顺铂化疗敏感性,分析胃癌组织中NOB1阳性表达情况,运用Logistic回归分析导致胃癌患者耐药的独立危险因素。结果胃癌组织NOB1蛋白表达与癌旁组织对比差异有统计学意义(P<0.05)。耐药组NOB1蛋白表达阳性率高于敏感组(P<0.05)。胃癌患者中顺铂敏感率为35.56%。耐药组肿瘤低、未分化,胃癌处于Ⅲ、Ⅳ期,NOB1阳性表达,10号染色体磷酸酶丢失者构成比均高于敏感组(P<0.05);敏感组服用抗肿瘤中药构成比高于耐药组(P<0.05);耐药组谷胱甘肽-S-转移酶>21U/L者构成比大于敏感组(P<0.05)。低、未分化,Ⅲ、Ⅳ期,NOB1蛋白阳性,10号染色体磷酸酶丢失,谷胱甘肽-S-转移酶>21U/L是胃癌患者发生耐药的独立危险因素(RO=4.289、2.098、7.142、1.527、1.488,P<0.05);服用抗肿瘤中药是胃癌组织发生耐药性的独立保护因素(OR=0.549,P<0.05)。结论NOB1在胃癌组织中表达上调,且与顺铂化疗敏感性有关,NOB1蛋白在胃癌组织中阳性表达可增加胃癌细胞的耐药性,是胃癌组织发生耐药性的独立危险因素。 Objective To explore the expression of NOB1 protein of yeast gene Noblp in gastric cancer tissues and the clinical relationship with the sensitivity of cisplatin chemotherapy.Methods In May 2017 to June 2019 the l 90 cases of gastric cancer patients undergoing radical gastrectomy at Hengshui Fifth People’s Hospital were selected as the research object.NOB1 protein expression in gastric cancer tissue and the tissue adjacent to carcinoma were detected by immunohistochemical SP method.The sensitivity of patients to cisplatin chemotherapy was evaluated.The positive expression of NOB1 in gastric cancer tissues of drugresistant group and sensitive group was analyzed,and the The component ratio of influencing factors affecting drug resistance in patients was calculated.The result of drug resistance in patients with gastric cancer risk factors were analyzed by Logistic regression.Results The positive rate of NOB1 protein in gastric cancer tissues was higher than that in adjacent tissues(P<0.05).The positive rate of NOB1 protein expression in the drug-resistant group was higher than that in the sensitive group(P<0.05).The sensitivity rate of cisplatin in patients with gastric cancer was 35.56%.The patients with gastric carcinoma of tumor,undifferentiated and gastric cancer in phaseⅢ,Ⅳ,NOB1 positive expression,chromosome 10 phosphatase of lost Low resistant group were higher than sensitive group(P<0.05),The composition ratio of taking anti-tumor TCM in the sensitive group was higher than in the resistant group(P<0.05).The composition ratio of glutathione-s-transferase>21 U/L was higher than that of the sensitive group(P<0.05).Low,undifferentiated,Ⅲ,Ⅳperiod,NOB1 protein positive expression,chromosome 10 phosphatase lost recurrence,GSH-S-transferase>21 U/L were risk factors of drug resistance in patients with gastric carcinoma(OR=4.289,2.098,7.142,1.527,1.488,P<0.05);Taking anti-tumor traditional Chinese medicine was the protective factor of drug resistance in gastric cancer tissues(OR=0.549,P<0.05).Conclusion The
