摘要
目的:探讨粉防己碱(TET)对胃癌耐药细胞锌指蛋白139(ZNF139)、多耐药基因1(MDR1)及谷胱甘肽S转移酶-π(GST-π)mRNA的影响。方法:选取胃癌细胞SGC7901以及胃癌耐阿霉素细胞SGC7901/ADR,采用逆转录-聚合酶链反应和蛋白质印迹法检测两种细胞ZNF139、MDR1、GST-πmRNA及蛋白表达,同时检测2.0!g·ml-1TET处理前后SGC7901/ADR细胞ZNF139、MDR1、GST-πmRNA及蛋白表达。结果:SGC7901/ADR细胞ZNF139、MDR1、GST-πmRNA相对表达量明显高于SGC7901细胞(P<0.05);SGC7901/ADR细胞ZNF139、MDR1、GST-π蛋白相对表达量明显高于SGC7901细胞(P<0.05);TET处理后SGC7901/ADR细胞ZNF139、MDR1、GST-πmRNA及蛋白相对表达量均较处理前明显降低(P<0.05)。结论:TET能下调胃癌耐药细胞ZNF139、MDR1、GST-πmRNA和蛋白表达水平,从而在逆转胃癌多药耐药特性方面有重要作用。
Objective: To investigate the effects of tetrandrine(TET) on zinc finger protein 139(ZNF139),multidrug resistance gene 1(MDR1) and glutathione S transferase-π(GST-π) mRNA of multidrug resistant gastric cancer cells. Methods: Gastric cancer cells SGC7901 and SGC7901/ADR were selected,and ZNF139,MDR1 and GST-π mRNA and protein expression in the two kinds of cells were detected by RT-PCR and Western blot.ZNF139,MDR1 and GST-π mRNA and protein expression of SGC7901/ADR cells were detected before and after2. 0 !g·ml-1 TET treatment. Results: ZNF139,MDR1 and GST-π mRNA relative expression of SGC7901/ADR cells were significantly higher than those of SGC7901 cells(P 〈 0. 05); ZNF139,MDR1 and GST-π protein relative expression of SGC7901/ADR cells were significantly higher than those of SGC7901 cells(P 〈 0. 05); After TET treatment ZNF139,MDR1 and GST-π mRNA and protein relative expression of SGC7901/ADR cells were significantly decreased compared with those before treatment(P 〈 0. 05). Conclusion: TET can down-regulate ZNF139,MDR1 and GST-π mRNA and protein expression in drug resistant gastric cancer cells,which plays an important role in reversing multidrug resistance characteristics of gastric cancer.
