Optimization of the alcohol extraction process of Compound Honghuang buccal tablets by Box-Behnken response surface method

摘要 目的优选复方红黄口含片醇提工艺。方法以红景天中的红景天苷和酪醇、黄芪中的毛蕊异黄酮和芒柄花素的含量及浸膏的得率为评价指标,以乙醇浓度、料液比、提取时间、提取次数等为考察因素,在单因素试验的基础上,对主要影响因素进行响应面分析,通过综合评分法优选最佳醇提工艺。结果优选得到的最佳醇提工艺为:乙醇浓度70%,料液比1∶14(g:m L),回流提取(138 min×2)。在此条件下,理论值与预测值接近,相对误差为0.90%。结论优选得到的复方红黄口含片醇提工艺稳定可行。 OBJECTIVE To optimize the alcohol extraction process of Compound Honghuang buccal tablets(CHBT).METHODS The contents of salidroside,p-tyrosol,calycosin,formononetin,and yields of the extracts were used as index,the ethanol concentration,material-liquid ratio,extraction time and frequency were used as the investigate factors for single factor experiment,and the Box-Behnken response surface method was used to analyze the main influence factors through the comprehensive scoring method to study the alcohol extracting technical.RESULTS The optimal extraction technology was as follows:Reflux extraction two times,each time 138 minutes.Ethanol concentration was 70%,and the material-liquid ratio was1∶14(g∶m L).Under these conditions,the observed value and predicted value was very close,and the deviation was 0.90%.CONCLUSION The optimized extraction technology of CHBT alcohol extraction process is stable and feasible.
