摘要 目的:选用果胶酶酶解法来优化苦李根多糖的提取工艺。方法:利用果胶酶在合适的水浴温度下提取苦李根多糖,通过单因素试验和正交试验选出提取苦李根多糖的最优组合,并通过稳定性、精密度和重复性试验来验证实验结果的准确性。结果:最终优化苦李根多糖提取条件为料液比为1∶25、酶解时间为105 min、果胶酶添加量为1.0%、酶解温度为30℃,在最优提取条件下苦李根多糖含量为59.28±0.01 mg/g。结论:利用酶解法可有效提取出苦李根多糖,通过单因素和正交试验确定了最佳的提取条件,为苦李根后续研究提供了一定的理论参考。 Objective: To optimize the extraction process of polysaccharide from Kuli radix distillate by pectinase. Methods: Pectinase was used to extract polysaccharide from Kuli radix distillate under suitable water bath temperature. The optimal combination of polysaccharides from Kuli radix distillate was selected by single factor test and orthogonal test, and the stability, precision and repeatability test were carried out to verify the accuracy of the experimental results. Results: The optimal extraction conditions of polysaccharides from Kuli radix distillate were as follows: the ratio of material to liquid was 1∶25, the time of enzymatic hydrolysis was 105 min, the amount of pectinase was 1.0%, and the temperature of enzymatic hydrolysis was 30 ℃. Under the optimal extraction conditions, the content of polysaccharides in Kuli radix distillate was 59.28±0.01 mg/g. Conclusion: Polysaccharide from Kuli radix distillate can be effectively extracted by enzymatic hydrolysis. The best extraction conditions were determined by single factor and orthogonal test, which provided a theoretical reference for the follow-up study of Kuli radix distillate.
