摘要
为探讨G蛋白偶联雌激素受体(GPER)在猪不同大小卵泡的分布与表达,分离直径小于3 mm、3~5 mm及大于5 mm健康完整有腔卵泡,分别采用免疫组化、蛋白免疫印迹及免疫荧光结合共聚焦成像等方法,对各组卵泡中GPER的分布及相对表达进行研究。结果表明,GPER在猪不同大小卵泡的颗粒细胞、膜细胞、卵丘细胞和卵母细胞质膜及胞质中均有分布,以大于5 mm卵泡组颗粒细胞分布最多、着色最深;GPER相对表达量随着卵泡的发育而逐渐升高。GPER在猪不同大小卵泡的分布和表达量变化与卵泡的发育过程大体一致,提示其可能介导了雌激素调节猪卵泡发育与卵母细胞成熟的过程。
In order to investigate the distribution and expression of the G protein coupled estrogen receptor(GPER)in different sizes of follicles in pigs,the follicles of diameters<3 mm,3-5 mm and>5 mm were isolated.Their distribution and relative expression were studied by immunohistochemistry,Western blotting and immunofluorescence combined with confocal imaging analysis,respectively.The results showed that GPER was widely located in the granular cells,theca cells,cumulus cells and oocytes plasma membranes and cytoplasms of different sizes of follicles in pigs,and the deepest staining of GPER was found in the granular cells of the>5 mm follicle group.The relative expression of GPER increased gradually along with follicular development.The distribution and expression of GPER in different sizes of follicles in pigs were generally consistent with that of follicular development,which might mediate the process of regulating follicular development and oocyte maturation in pigs by estrogen.
