摘要
[目的]观察白介素1β(IL-1β)在大鼠卵泡发育过程中的表达以及功血宁Ⅱ号冲剂对其表达的影响,探讨该药促进卵泡发育诱发排卵的作用机制。[方法]将未成年雌性大鼠随机分为模型对照组、阳性对照组和功血宁Ⅱ号高、低剂量组,分别给予蒸馏水、安坤赞育丸和功血宁Ⅱ号冲剂,14d后皮下注射妊马血清促性腺激素(PMSG)+人绒毛膜促性腺激素(hCG)制备诱发排卵大鼠的模型。分别在卵泡发育期和排卵前期处死动物摘取卵巢,常规制备卵巢切片,免疫组化法检测各组动物卵泡不同发育时期IL-1β表达。[结果]IL-1β在模型对照组中的表达,排卵前期的阳性细胞面积、阳性面积率、积分光密度、平均光密度均显著高于卵泡发育期(P<0.05);功血宁Ⅱ号组在卵泡发育期和排卵前期IL-1β表达的上述指标均明显高于模型对照组(P<0.05或P<0.01),排卵前期的上述指标均明显高于相应卵泡发育期(P<0.05或P<0.01),其中高剂量组的作用较低剂量组和阳性对照组显著。[结论]IL-1β在大鼠卵泡发育过程中均有表达并逐渐增强。功血宁Ⅱ号冲剂能够显著提高IL-1β的表达,通过其参与卵母细胞成熟,诱导卵巢排卵因子的产生,在卵泡破裂激发排卵过程中发挥作用,促进卵泡发育诱发排卵。
[Objective] To observe the expression of interleukin 1β(IL-1β) during the development of follicle and the effect of GongXueNing Chongji No.Ⅱ (Stopping the hemorrhage from the uterus) on it. [Methods] immature female rats were randomly divided into model control group, positive control group, GongXueNing Ⅱ high and low dosage group. They were treated with distilled water, Ankunzanyu Pill and GongXueNing Chongji No.Ⅱ (taken medicine after infused in boiling water) respectively. 14 days later a model of ovulation in rats was induced by subcutaneous FMSG+hCG. The animals were sacrificed at development period of follicle and preovulatory period and their ovaries were harvested. The expression of IL-1β of pathologic section in the different development periods was measured with immunohistochemistry. [Results] In the model group the IL-1β expression, the positive cell area, the rate of positive area, the integrated optical density, the average optical density in preovulatory period were obviously higher than that in the development period of follicles (P〈0.05). In GongXueNing No.Ⅱ group the above parameters of IL-1β expression at the development period of follicles and preovulatory period were all higher than the model control group (P〈0.05 or P〈0.01). All the parameters described above at preovulatory period were obviously higher than that at the corresponding period of follicle development (P〈0.05 or P〈0.01). Among them, the effect of high dosage group was more obvious than that in low dosage group and the positive control group. [Conclusion] IL-1βwas expressed and increased gradually during the process of follicle development. GongXue Ning Chongji No. Ⅱ can significantly increase the expression of IL-1βand can accelerate the development of follicle and induce the ovulation.
