摘要
目的:探讨内服三黄泻心汤外用冰硼散对口腔溃疡(RAU)模型大鼠的治疗作用及对炎性介质的影响,探讨其作用机制。方法:50只健康SD大鼠,随机分为正常组(A)、模型组(B)、冰硼散组(C)、三黄泻心汤组(D)、冰硼散+三黄泻心汤组(C+D),每组10只。除正常组(A组)外,其余组大鼠均采用化学烧灼法在其右侧口腔黏膜建立RAU模型。模型成功建立后,大鼠分别以相应药物涂抹溃疡处,并给予相应药物灌胃,连续给药6天;正常组和模型组不作处理。观察给药第1天和第3天时大鼠溃疡面情况并测定溃疡面积;取部分溃疡组织切片观察其组织形态学变化;检测大鼠血清中白细胞介素2(IL-2)、白细胞介素10(IL-10)和肿瘤坏死因子α(TNF-α)的水平。结果:与空白组比较,模型组大鼠口腔溃疡明显,组织病变明显;血清中TNF-α、IL-2、IL-10水平均显著升高(P<0.05);与模型组比较,6 d后各给药组大鼠的溃疡面积均显著缩小(P<0.05)或几近愈合,组织病变明显减轻;血清中TNF-α、IL-2、IL-10水平均显著降低(P<0.05)。结论:冰硼散联合三黄泻心汤(内服+外用)能显著改善RAU模型大鼠溃疡症状,具有强效的抑菌、消肿、修复作用,其机制可能与降低炎症因子水平有关。
Objective:To investigate the therapeutic effect of oral administration and external application on rat model of oral ulcer and the action mechanism.Methods:50 healthy SD rats were randomly divided into the normal group,the model group,Bingpeng Powder group,Sanhuang Xiexin Decoction group,and the combination group(Bingpeng Powder+Sanhuang xiexin Decoction),with 10 rats in each group.Oral ulcer model was established by chemical burning to the right oral mucosa in all groups except the normal group.After successful modeling,corresponding drugs were smeared to the ulcer site and cornesponding oral administration was applied for 6 days in the medication groups.The ulcer surface was observed and the ulcer area was measured on the 1st and 3rd days after medication.Some ulcer tissue sections were taken to observe the histomorphological changes.The serum levels of IL-2,IL-10 and TNF-αwere measured.Results:The oral ulcer and histopathological changes were obvious,and the serum levels of TNF-α,IL-2 and IL-10 were significantly increased in the model group compared to those in the normal group(P<0.05).The ulcer area was significantly reduced or nearly healed(P<0.05);the histopathological changes were significantly reduced;the serum levels of TNF-α,IL-2 and IL-10 were significantly decreased in all medication groups compared to those in the model group(P<0.05).Conclusion:Bingpeng Powder combined with Sanhuang Xiexin Decoction(oral administration+external application)can significantly improve the ulcer symptoms of the model rats,and the therapy has strong effects of bacteriostasis,detumescence and repair.Its mechanism may be related to the reduction of inflammatory factors.
