摘要
目的:观察通窍止鼽汤对过敏性鼻炎大鼠的作用,并观察其对免疫功能指标的影响。方法:随机将大鼠分为正常对照组(10只)、模型组(10只)以及中药组(21只)。模型组及中药组大鼠建立过敏性鼻炎模型,中药组随机分为通窍止鼽汤高剂量组、通窍止鼽汤中剂量组及通窍止鼽汤低剂量组,每组7只,每日1次;对照组及模型组大鼠接受等剂量生理盐水灌胃,每日1次;连续干预10d,疗程结束后观察各组大鼠鼻部继发症状的变化,同时分别取鼻黏膜组织及血清,用HE染色法观察鼻黏膜组织的病理变化,用ELISA检测血清组胺(HIS)、免疫球蛋白E(IgE)、白介素-4(IL-4)、肿瘤坏死因子-α(TNF-α)的浓度变化。结果:(1)与正常对照组比较,模型组及中药组大鼠出现明显的打喷嚏,鼻部红肿和挠鼻症状,其中中药组较模型组有所改善,并其中高剂量组及中剂量组改善较低剂量组明显,差异均有统计学意义(P<0.05),高剂量组与中剂量组之间差异无统计学意义(P>0.05);(2)空白对照组大鼠鼻粘膜结构完整,上皮细胞排列整齐,平滑,PH无炎症表现;模型组大鼠鼻粘膜出现大量上皮细胞脱落、坏死,出现大量的嗜酸性粒细胞浸润,杯状细胞增生,粘膜表面出现明显的分泌物;中药组大鼠鼻粘膜较模型组改善,炎性浸润征象有所缓解,其中高剂量组及中剂量组改善较低剂量组明显,高剂量组与中剂量组之间形态相近。(3)与空白组比较,模型组及中药组大鼠血清HIS、 IgE、IL-4及TNF-α均明显升高,其中中药组较模型组浓度下调,其中高剂量组及中剂量组改善较低剂量组明显,差异均有统计学意义(P<0.05),高剂量组与中剂量组之间差异无统计学意义(P>0.05)。结论:通窍止鼽汤可明显改善过敏性鼻炎症状及组织病理变化,其作用机制可能通过抑制免疫因子释放有关。
Objective:To observe the effect of Tongqiao Zhirong Decoction on allergic rhinitis rats and its influence on im-mune function.Methods:The rats were randomly divided into normal control group(10rats),model group(10rats)and Chinese medicine group(21rats).Rats in model group and Chinese medicine group were randomly divided into three groups:high-dose Tongqiao Zhirong Decoction group,middle-dose Tongqiao Zhirong Decoction group and low-dose Tongqiao Zhirong Decoction group,7rats in each group,once a day;rats in control group and model group received the same dose of normal saline,once a day;rats in control group and model group received the same dose of saline,once a day;after 10days of continuous intervention,after the end of the course of treatment.The pathological changes of nasal mucosa and serum were observed by HE staining.The concentrations of histamine(HIS),immunoglobulin E(IgE),interleukin-4(IL-4)and tumor necrosis factor-alpha(TNF-alpha)in serum were detected by ELISA.Results:(1)Compared with the normal control group,the model group and the tradi-tional Chinese medicine group showed obvious sneezing,nose swelling and nasal scratching symptoms.The traditional Chinese medicine group was better than the model group,and the high-dose group and the middle-dose group improved significantly in the low-dose group,the difference was statistically significant(P<0.05),the high-dose group and the traditional Chinese medicine group improved significantly(P<0.05).There was no significant difference between the two groups(P>0.05);(2)The structure of nasal mucosa in the blank control group was intact,the epithelial cells were arranged neatly and smoothly,and there was no inflammation;the nasal mucosa in the model group showed a large number of epithelial cells exfoliation,necrosis,eosinophil infiltration,goblet cell proliferation,mucosal surface.The nasal mucosa of rats in the traditional Chinese medicine group was improved and the inflammatory infiltration was alleviated.The high dose group and the middle dose group impro
