摘要
目的:探讨黄连素(BBR)对大鼠结肠敏感性和动力学异常引起肠易激综合征(IBS)的影响及其机制。方法:选取SPF级成年SD雄性大鼠,分为对照组、IBS组和BBR干预组(BBR组),各15只。IBS组和BBR组应用直结肠扩张术(CRD)建立IBS模型,造模成功后,BBR组大鼠按3ml/次/天BBR灌胃,1个疗程/7天,连续2个疗程。灌胃开始至4周后第2天,观察各组大鼠的体质、排便、结直肠敏感性和动力异常的主要指标变化;酶解法分离三组大鼠单个结肠平滑肌细胞,全细胞膜片钳技术记录各组结肠平滑肌细胞膜上ICa-L的电流峰值及其电流-电压曲线。结果:与IBS组比较,BBR组大鼠体质好转,排便次数减少,体重增加更明显,2h内排出粪粒数及其含水量均显著减少,引起IBS发生率亦降低(P<0.01),基本接近对照组水平(P>0.05);CRD实验结果显示,与IBS组比较,BBR组大鼠腹部抬起和背部拱起所需注水量增加,结肠运动指数及其变化率减小(P<0.01);全血胞膜片钳技术结果显示,与IBS组比较,BBR组大鼠结肠平滑肌细胞ICa-L幅度较小,且随着钳制电压增大ICa-L减小幅度更明显。当钳制电位为+10mv时,BBR组大鼠结肠平滑肌细胞ICa-L明显小于IBS组(P<0.01)。BBR组大鼠结肠平滑肌细胞ICa-L的I-V曲线较IBS组明显上抬,接近对照组。结论:BBR能明显改善IBS的临床症状及结肠的高敏感性和动力异常,且作用持久不易复发。其机制可能与BBR抑制了IBS大鼠结肠平滑肌细胞ICa-L,改良结肠平滑肌细胞离子通道电重构有关。
Objective:To investigate the effects and mechanism of berberine(BBR)on irritable bowel syndrome(IBS)induced by visceral hypersensitivity and abnormality in rats.Method:The male Sprauge-Dawley rats with SPF type were divided into normal control,IBS and BBR group(each group n=15).The model about IBS was established via using colorectal distension method(1ones/d for 1week),after justified successfully by changes of parameters of clinical symptoms with bowel evacuation habit,characteristics and quantity in rats.The rats about BBR group came from these of the IBS rats to lavage continuously with BBR extracting solution(3ml/day)in 14 days.After that to evaluate the effects of BBR administration on IBS,some parameters or scores were measured and statistics through clinical symptoms such as visceral hypersensitivity and abnormality as well colorectal distension(CRD)in rats of three groups.To dissociate enzymatically single rat colonic myocytes and to record L-type calcium current(ICa-L)and its current-voltage(I-V)curve via whole-cell patch clamp technique,the mechanism was studied about treatment of BBR on IBS in rats.Results:Compared with these in IBS group,the clinical symptoms were all improved,such as the fecal particles were become more dry,and the body and food weight but nor water were significantly increased,and the fecal particles and its water weight discharged in 2hours were obviously decreased,as well as incidence rate of IBS was severely decreased from 93%in IBS group to 33%in rats of BBR groups(Pof all<0.01,respectively).Moreover,the water injection rates with abdominal withdrawal reflex(AWR)were completely increased,as well as the colonic motility index and its change rate were significantly decreased via CRD compared with IBS group in IBS rats administrated with BBR(Pof all<0.01,respectively).Finally,ICa-Lwas significantly decreased come from different membrane voltage,and the I-V curve was notably change upward so approach to control group in colonic cells of rats in BBR group but not IBS group.With volt
