黄连素对脊髓损伤大鼠BMP-4 mRNA表达及神经元活性的 影响 被引量:1

Effects of berberine on BMP-4 mRNA expression and neuronal activity in rats with spinal cord injury

摘要 目的研究黄连素(BBR)对脊髓损伤大鼠HE病理、骨形态发生蛋白4(BMP-4)mRNA表达及神经元活性的影响。方法将在广东省医学SD大鼠中心的大鼠随机分为脊髓损伤组(S组);健康对照组(H组);黄连素组(B组),每组10只。采用流式细胞法、TUNEL法、HE染色法免疫组化法、qRT-PCR法检测S组、B组、H组大鼠脊髓细胞凋亡数量、脊髓细胞病变程度、脊髓细胞中BMP-4蛋白阳性表达、脊髓细胞中BMP-4 mRNA的表达含量、脊髓细胞凋亡率的差异性来探究BBR对脊髓损伤大鼠HE病理、BMP-4 mRNA表达及神经元活性的影响。结果S组脊髓细胞凋亡数量显著多于B组(P<0.05),与H组相比较,B组脊髓细胞凋亡数量显著多于H组(P<0.05)。B组神经元细胞的凋亡情况与S组相比较显著减少(P<0.05)。与S组比较,B组大鼠脊髓水肿减轻,损伤程度有好转趋势,细胞坏死数目有减少。与S组相比较,B组脊髓细胞中BMP-4阳性数显著降低(P<0.05)。与B相比较,H组细胞BMP-4阳性数显著下降(P<0.05);与S组相比较,B组大鼠BMP-4 mRNA表达含量显著降低,BMP-2 mRNA表达量显著上升(P均<0.05),与B组相比较,H组BMP-4 mRNA表达含量显著降低,BMP-2 mRNA表达量显著上升(P均<0.05)。结论黄连素(BBR)对脊髓损伤大鼠具有一定治疗作用,BBR干预后能够显著减少大鼠脊髓细胞的凋亡数量,降低凋亡率,同时,改善大鼠脊髓细胞的病理形态,促进康复,BBR对脊髓损伤大鼠的作用机制可能与下调脊髓细胞中BMP-4的表达有关。 Objective To study the effects of berberine(BBR)on HE pathology,bone morphogenetic protein 4(BMP-4)mRNA expression and neuronal activity in rats with spinal cord injury.Methods The rats in the Guangdong Medical SD Rat Center were randomly divided into spinal cord injury group(S group);healthy control group(H group);berberine group(B group),each group with 10 rats.Flow cytometry,TUNEL method,HE staining method,immunohistochemical method,and qRT-PCR method were used to detect the apoptosis number,the degree of spinal cord cytopathy,the expression level of BMP-4 protein and mRNA,and the apoptosis rate of spinal cord cells for the analysis of the effects of BBR on HE pathology,BMP-4 mRNA expression and neuronal activity in rats with spinal cord injury.Results The number of spinal cord apoptosis in group S was significantly higher than that in group B(P<0.05).Compared with group H,the number of spinal cord apoptosis in group B was significantly higher than that in group H(P<0.05).The apoptosis of neuronal cells in group B was significantly reduced compared with group S(P<0.05).Compared with the S group,the spinal cord edema of the rats in the B group was reduced,the degree of injury tended to improve,and the number of cell necrosis decreased.Compared with group S,the number of BMP-4 positive in spinal cord cells of group B was significantly reduced(P<0.05).Compared with group B,the number of positive BMP-4 cells in group H decreased significantly(P<0.05);compared with group S,the expression of BMP-4 mRNA in group B rats significantly decreased,and the expression of BMP-2 mRNA increased significantly(P<0.05).Compared with group B,the expression of BMP-4 mRNA in group H was significantly reduced,and the expression of BMP-2 mRNA was significantly increased(P<0.05).Conclusion Berberine has a certain therapeutic effect on rats with spinal cord injury.After BBR intervention,it can significantly reduce the number of rat spinal cord apoptosis.At the same time,it can improve the pathological morphology of rat spinal cord cells
