摘要
以红菜苔的根、茎、叶、花、种子为试材,用MS+6-BA 0.5 mg·L^-1+2,4-D 0.5 mg·L^-1+NAA 0.8 mg·L^-1改良培养基诱导红菜苔不同外植体的愈伤组织,根据其诱导情况筛选出最佳外植体;通过单因素和响应面试验法,研究了6-BA、2,4-D和NAA 3种植物生长调节剂对红菜苔最佳外植体愈伤组织诱导的影响,进一步优化红菜苔愈伤组织的诱导条件;使用不同抗褐化剂研究其对红菜苔茎愈伤组织诱导情况的影响,以期筛选出能有效防止或减轻愈伤组织褐化,提高愈伤组织诱导率和生长率的最佳抗褐化剂。结果表明:红菜苔茎是诱导愈伤组织的最佳外植体,其诱导率为93.33%;茎愈伤组织诱导的最佳培养基为MS+6-BA 0.6 mg·L^-1+2,4-D 0.8 mg·L^-1+NAA 1.0 mg·L^-1,诱导率达到94.2%,且大多数愈伤组织状态及长势良好;0.2 g·L^-1的活性炭是红菜苔茎愈伤组织诱导的最佳抗褐化剂,褐化率为23.4%且愈伤组织生长状况良好,可以为开展红菜苔的离体再生体系提供参考依据。
The callus was induced by the modified medium of MS+6-BA 0.5 mg·L^-1+2,4-D 0.5 mg·L^-1+NAA 0.8 mg·L^-1 with the root segment,stem segment,leaf,flower and seed of red cabbage,and the best explant was screened according to its induction.By single factor and response surface test,the effects of three plant growth regulators 6-BA,2,4-D and NAA on callus induction of the best explants of red cabbage were studied.The effect of callus induction further was optimized the conditions for callus induction of red cabbage.The effects of different anti-browning agents on the callus induction of red cabbage were studied to screen out the best anti-browning agent which can effectively prevent or reduce the browning of callus and improve the callus induction rate and growth rate.The results showed that the stem was the best explant of callus induced by red cabbage,the induction rate of stem was 93.33%.The optimum concentration of 6-BA,2,4-D,NAA were 0.6 mol·L^-1,0.8 mol·L^-1,1.0 mol·L^-1.The induction rate was 94.2%.It was suitable for the callus induced by the callus of the stem segment of the red cabbage:MS+6-BA 0.6 mg·L^-1+2,4-D 0.8 mg·L^-1+NAA 1.0 mg·L^-1;0.2 g·L^-1 AC was the best anti-browning agent induced by callus of red cabbage,the browning rate was 23.4% and the callus grows well.The best explants induced by callus of red cabbage and the optimization process were obtained in this experiment,it could provide a theoretical basis for the in vitro regeneration system of red cabbage.
