摘要
以黄果枸杞无菌苗的叶片和茎段为外植体,研究不同激素配比的培养基对外植体愈伤组织的诱导、丛生芽的分化及生根培养的影响,获得再生植株,筛选出适宜各阶段培养的最佳培养基配方,从而建立黄果枸杞组织培养和快繁技术新体系。结果表明:适合嫩茎段腋芽诱导与生长的培养基为MS;叶片和茎段都适合黄果枸杞愈伤组织的诱导,愈伤组织诱导培养基为MS+6-BA 0.4-0.6mg/L+NAA1.0mg/L,诱导率为90.23%和89.14%;诱导愈伤组织分化培养基为MS+6-BA 0.6mg/L+IBA 0.15mg/L,分化率90.12%;诱导丛生芽生根适宜培养基为1/2MS+NAA0.4mg/L+IBA0.6mg/L,生根率为90.33%;诱导生根的组培苗在实验室内打开瓶口炼苗7d,在温室内移栽到适宜的营养钵中,营养钵中的基质为森林土、河沙及蛭石的比例为2:1:1,移栽成活率达93%。
Using the leaf and stem section of Lycium barbarum L. var. auranticarpum as explants, the effect of the medium with different hormone combination on callus induction and the differentiation of multiple shoot and rooting was researched, in order to select the best culture medium formula suitable for each stage and establish the tissue culture and rapid propagation system. The results showed that MS medium was fit for axillary bud induction and growth of tender stem segments. Leaf and stem sections were all the suitable explants for callus induction. Callus induction medium was MS + 6 - BA 0.4 -0.6mg/L + NAA1.0mg/L, the callus induction rate was 90. 23% and 89.14%, respectively. The callus differentiation induction media was MS +6 - BA 0.6mg/L + IBA 0. 15mg/L and differentiation rate was 90.12%. Rooting medium was 1/2MS + NAA0.4mg/L + IBA0.6mg/L and the rooting rate was 90.33%. After hardening - seedling of the rooting plantlets for 7 days, the seedlings were transplanted in feeding block with the matrix including forest soil, sand and vermiculite under the proportion of 2 : 1 : 1, and the survival rate of seedling reached 93 %.
