摘要
【目的】考察不同剂量女黄颗粒对免疫抑制小鼠外周血T淋巴细胞亚群及细胞因子IL-2、IL-4、TNF-α和IFN-γ表达量的影响,旨在明确女黄颗粒对小鼠免疫的调节作用。【方法】选取50只健康昆明小鼠,随机分为阴性对照组、模型组及女黄颗粒高、中、低剂量组,每组10只。除阴性对照组外,其他处理组均通过腹腔注射环磷酰胺(80 mg/kg,1次/d,连续5 d)建立小鼠免疫抑制模型;建模后,阴性对照组和模型组灌胃生理盐水,女黄颗粒处理组则分别按0.5、1.0和1.5 g/kg剂量灌胃女黄粒颗溶液(以水稀释),1次/d,连续7 d。采用流式细胞仪检测小鼠外周血T淋巴细胞亚群CD3+、CD4+和CD8+的百分率及CD4+/CD8+比值,以实时荧光定量PCR检测小鼠脾脏细胞因子IL-2、IL-4、TNF-α和IFN-γ的表达情况。【结果】经腹腔注射环磷酰胺后,小鼠的CD3+、CD4+和CD8+T淋巴细胞百分率及CD4+/CD8+比值较阴性对照组小鼠极显著降低(P<0.01,下同),小鼠脾脏IL-2、IL-4、TNF-α和IFN-γmRNA的相对表达量极显著下降。灌胃给药(女黄颗粒)后,CD3+、CD4+和CD8+T淋巴细胞百分率及CD4+/CD8+比值均有所升高,其中女黄颗粒高、中剂量组小鼠的CD3+、CD4+和CD8+T淋巴细胞百分率较模型组小鼠极显著升高;除女黄颗粒高剂量组小鼠脾脏IL-2 mRNA的相对表量较模型组小鼠极显著下降外,各女黄颗粒处理组小鼠脾脏IL-2、IL-4、TNF-α和IFN-γmRNA的相对表达量均呈上升趋势,尤其是女黄颗粒中剂量组小鼠脾脏IL-4、TNF-α和IFN-γmRNA的相对表达量均极显著高于模型组小鼠。【结论】女黄颗粒通过上调CD3+、CD4+、CD8+T淋巴细胞百分率及提高IL-2、IL-4、TNF-α、IFN-γ的相对表达量以增强免疫抑制小鼠的免疫功能,临床推荐使用剂量为1.0 g/kg。
【Objective】The effects of Nühuang granules of different doses on expression of peripheral blood T lymphocyte subsets and cytokines IL-2,IL-4,TNF-αand IFN-γin immunosuppressed mice were investigated to clarify the regulation of Nühuang granules on mouse immunity.【Method】Fifty healthy Kunming mice were randomly divided into negative control group,model group,and Nühuang granules high-,mid-,and low-dose groups,with ten mice in each group.Except for the negative control group,the other groups were treated with intraperitoneal injection of cyclophosphamide(80 mg/kg BW,once daily for 5 d)to establish an immunosuppressive model.After successful modeling,the negative control group and the model group were given normal saline by gavage,and the Nühuang granules groups was administered with 0.5,1.0 and 1.5 g/kg doses(diluted with water)once a day for 7 d by gavage.Simultaneously,the percentages of CD3^+,CD4^+,CD8^+and CD4^+/CD8+ratios of peripheral blood T lymphocyte subsets were detected using flow cytometry,and the expression of cytokines IL-2,IL-4,TNF-α,IFN-γin spleen was detected by real-time fluorescence quantitative RT-PCR.【Result】After injected with cyclophosphamide,the percentages of CD3^+,CD4^+,CD8^+T lymphocyte and the ratio of CD4^+/CD8^+in mice were extremely reduced(P<0.01,the same below),and the relative expressions of IL-2,IL-4,TNF-α,and IFN-γmRNA were extremely decreased.After administration of Nühuang granules by gavage,the percentage of CD3^+,CD4^+and CD8^+T lymphocytes and the ratio of CD4^+/CD8^+were increased.Among them,the percentages of CD3^+,CD4^+and CD8^+T lymphocytes in the Nühuang granules high-and mid-dose groups were extremely increased.The relative expression of IL-2 mRNA in the spleen of Nühuang granules high-dose group was extremely decreased.But relative expression levels of IL-2、IL-4,TNF-αand IFN-γin the spleens in other treatment groups increased,especially the expression level of IL-4,TNF-αand IFN-γin the spleen of Nühuang granules mid-dose group was extrem
