摘要
以总黄酮含量和干浸膏得率为考察指标,采用单因素试验和L9(34)正交试验优化辣木叶提取物的制备工艺,并通过环磷酰胺造模法复制实验小鼠免疫抑制模型,探讨辣木叶提取物对实验小鼠免疫功能调节的影响。结果表明,辣木叶提取物的优选制备工艺参数为:以70%乙醇提取,料液比1∶15,提取温度80℃,提取3次,每次1.5 h。此工艺条件下的辣木提取物的总黄酮质量分数为15.64 mg·g-1,能显著提高免疫器官指数,增强单核吞噬细胞功能,促进血清免疫球蛋白Ig G和溶血素的合成,降低血清AST活性,对免疫功能抑制有调节作用。
With total flavonoid content and dry extract yield as the observation indexes,the optimal extraction conditions of Moringa oleifera leaves were determined by using single factor test and orthogonal test,and cyclophosphamide modeling method was used to establish immunosuppressed mice models,so as to investigate the effects of M. oleifera leaves extract on immune regulation in mice. The results showed that the optimal preparation conditions were as follows: extraction with 70% ethanol,material-liquid ratio 1∶ 15,extraction temperature 80 ℃,three times,1. 5 hours for each time. Under these conditions,the content of total flavonoids from M. oleifera leaves was 15. 64 mg·g-1,which can significantly enhance macrophage phagocytosis and immune organ index,promote the synthesis of serum immunoglobulin Ig G and hemolysin,and decrease AST activity,with regulation effect on immune dysfunction.
