摘要
本研究旨在深入探讨沙冬青种子总黄酮对免疫抑制小鼠的免疫功能及体外脾淋巴细胞增殖的影响。试验分体内与体外两部分。体内试验:将100只昆明种小鼠随机分为5组,每组20只(雌雄各半)小鼠,分别为空白对照组(生理盐水组)、免疫抑制模型组、总黄酮灌胃试验Ⅰ组[50 mg/(kg·d)]、试验Ⅱ组[150mg/(kg·d)]和试验Ⅲ组[250 mg/(kg·d)]。上述小鼠试验28 d后,分别测定碳粒廓清指数和脏器指数(胸腺、脾)及小鼠血清IL-2、IL-4含量。体外试验:制备小鼠脾淋巴细胞悬液后,试验分为4组:沙冬青种子总黄酮作用组(试验组Ⅰ)、沙冬青种子总黄酮+Con A混合组(试验组Ⅱ)、Con A阳性对照组(试验组Ⅲ)、细胞悬液阴性对照组(试验组Ⅳ)。采用体外细胞培养技术、噻唑蓝(MTT)还原法检测体外培养脾淋巴细胞增殖的变化。结果表明,灌胃环磷酰胺可复制小鼠免疫抑制模型。不同剂量的沙冬青种子总黄酮能不同程度地提高免疫抑制小鼠胸腺和脾指数、碳粒廓清指数和吞噬指数以及血清IL-2和IL-4含量,尤其总黄酮灌胃剂量为150mg/(kg·d)的试验Ⅱ组,可显著提高免疫抑制小鼠的胸腺和脾指数、碳粒廓清指数和吞噬指数以及血清IL-2和IL-4含量,与免疫抑制模型组比,差异极显著(P〈0.01)。同时,沙冬青种子总黄酮可显著诱导小鼠脾淋巴细胞体外增殖,且这种诱导增殖效应与沙冬青种子总黄酮作用剂量呈正相关性。此外,Con A可协同沙冬青种子总黄酮诱导体外培养脾淋巴细胞的增殖。上述结果提示,沙冬青种子总黄酮可通过促进免疫器官淋巴细胞的增殖和成熟,改善和增强免疫抑制小鼠的免疫功能。
To investigate the effect of total flavonoids from the seeds of Ammopiptanthus mongolicus on the immune function in immunosuppressed mice and the proliferation of splenic lymphocyte of mice,some experiments were carried out in vivo and in vitro.In vivo experiments,100 Kunming mice were randomly divided into five groups(n=20,half male and half female):normal control group,model group and test groupsⅠ,Ⅱ,Ⅲ.The concentration of total flavonoids in the test groupsⅠ,Ⅱ and Ⅲ are 50 mg/(kg·d), 150 mg/(kg·d) and 250 mg/(kg·d),respectively.After 28 days,carbon clearance index,thymus index,spleen index,and the contents of IL-2 and IL-4 in serum were detected.In vitro experiments,the splenic lymphocyte of mouse were prepared and used as target cell,and divided into four groups:test groupⅠ,test groupⅡ,test group Ⅲ and test group Ⅳ.The effects were measured by MTT.In result,the immunosuppressed mice were copied successfully with CTX,and the total flavonoids from the seeds of Ammopiptanthus mongolicus increased the carbon clearance index,thymus index,spleen index,and the contents of IL-2 and IL-4 in serum in the immunosuppressed mice.The difference were significant compared with the model group(P〈0.01),especially in the test groupⅡ[150 mg/(kg·d)total flavonoids].At the same time,the total flavonoids from the seeds of Ammopiptanthus mongolicus was able to induce obviously proliferation of splenic lymphocytes of mice in vitro,and the effects with dependence of dosage,and Con A could increase the effects.The results suggest that the total flavonoids from the seeds of Ammopiptanthus mongolicus can reverse and repair obviously the immune function of the immunosuppressed mice and promote obviously the proliferation of splenic lymphocytes of mice.
