不同产地石上柏中3种双黄酮含量测定 被引量:4

摘要 目的:建立HPLC-DAD法同时测定石上柏中穗花杉双黄酮、扁柏双黄酮、银杏双黄酮的含量。方法:色谱柱:DikmaDiamons C18柱(150 mm×4.6 mm,5μm);流动相:乙腈-0.1%甲酸水,梯度洗脱;检测波长:330 nm;柱温:25℃;流速:0.6 mL/min;进样量:10μL。结果:3种双黄酮在上述色谱条件下分离度良好,穗花杉双黄酮的线性范围为0.68~4.08 mg/mL,标准曲线为Y=5293X-2881.8(R2=0.9995);扁柏双黄酮的线性范围为0.45~2.69 mg/mL,标准曲线为Y=1052.1X-242.58(R2=0.9996);银杏双黄酮的线性范围为0.70~4.13 mg/mL,标准曲线为Y=1513X-1441.6(R2=0.9995)。方法学考察,精密度、稳定性RSD〈2.0%,穗花杉双黄酮、扁柏双黄酮和银杏双黄酮的平均加样回收率分别为99.03%、97.65%、93.16%,(RSD〈3.0%)。10个产地石上柏中穗花杉双黄酮的含量在0.232%~0.309%,扁柏双黄酮的含量在0.040%~0.137%,银杏双黄酮的含量在0.043%~0.074%。结论:不同产地石上柏的3种双黄酮化合物的含量有一定的差异,可能与不同地域,不同采收时间以及不同的加工方法有关。该方法操作简便,测定准确,可用于中药石上柏的质量分析。 Objective:To establish a HPLC-DAD method for the simultaneous determination of amentoflavone,hinokiflavone,ginkgetin in Selaginella doederleinii. Methods:The chromatographic separation was performed on a DikmaDiamons C18 column(150 mm×4.6 mm,5 μm),adopting acetonitrile-0.1% formic acid solution as the mobile phase,and the detection wavelength at 330 nm,the column temperature was set at 25 ℃ and the flow rate was 0.6 mL/min,the injection volume was 10 μL. Results:This method had good linear relationship in range of 3.1-15.6 mg/mL and standard curve line of Y=5293 X-2881.8(R2=0.9995)for amentoflavone,1.1-5.6 mg/mL and standard curve line of Y=1052.1 X-242.58(R2=0.9996)for hinokiflavone and 0.7-3.4 mg/mL and standard curve line of Y=1513 X-1441.6(R2=0.9995)for ginkgetin,the RSD of precision and repeatability were less than 2.0%,the average recoveries(n=6)of amentoflavone,hinokiflavone and ginkgetin were 99.03%,97.65%,93.16%(RSD〈3.0%). The content range of amentoflavone was 0.232%-0.309%,the content range of hinokiflavone was 0.040%-0.137%,and the content range of ginkgo was 0.043%-0.074%. Conclusion:There are some differences on the content of three bioflavonoids from different habitats,which may be related to different regions,different harvest times and different processing methods.
