Nrf2介导姜黄素对四氯化碳诱导鲫鱼肝损伤的保护作用 被引量:6

Nrf2 mediate protective effects of curcumin against carbon tetrachloride (CCl_4)-induced liver injury in Carassius auratus

摘要 为了评价姜黄素对四氯化碳(CCl_4)诱导鲫鱼肝损伤的保护作用以及核因子NF-E2相关因子(Nrf2)的调控作用,采用精密肝切片技术(Precision-cut liver slices,PCLS)分离肝组织并进行体外培养,以四氯化碳诱导建立急性肝损伤模型,同时用不同浓度(1μg/ml、5μg/ml和10μg/ml)的姜黄素处理肝组织切片,收集上清液及肝组织。检测谷丙转氨酶(GPT)、谷草转氨酶(GOT)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、碱性磷酸酶(AKP)活性,以及丙二醛(MDA)、谷胱甘肽(GSH)、总蛋白(TP)、白蛋白(Alb)含量和总抗氧化能力(T-AOC)等生化指标,同时检测Nrf2和Keap1基因表达。结果表明:姜黄素可以改善CCl_4诱导的肝损伤,其浓度为1μg/ml和5μg/ml时能有效抑制GPT、GOT、LDH和AKP活性的升高,提高SOD、GSH-Px活性及T-AOC和GSH含量,降低MDA的生成,上调Nrf2基因表达。可见,姜黄素对CCl_4诱导的鲫鱼肝损伤有保护的作用,其作用机制可能是姜黄素能激活Nrf2基因在肝细胞中的表达,调节氧化应激,从而减轻CCl_4对肝损伤。 To evaluate the protective effects of curcumin on liver injury induced by carbon tetrachloride( CCl_4) and the regulatory role of nuclear factor NF-E2 related factor( Nrf2) in Carassius auratus,the liver tissue was isolated by precision-cut liver slices( PCLS) and cultured in vitro. The PCLS was treated with carbon tetrachlorideand curcumin( 1 μg/ml,5 μg/ml and 10 μg/ml). Then the culture supernatants and slices were collected for the analyses of biochemical parameters,such as activities of glutamatepyruvate transaminase( GPT),glutamate oxalatetransaminase( GOT),lactate dehydrogenase( LDH), superoxide dismutase( SOD), glutathione peroxidase( GSH-Px), catalase( CAT) and alkaline phosphatase( AKP),and contents of malondialdehyde( MDA),glutathione( GSH),total protein( TP),albumin( Alb),and total antioxidant capacity( T-AOC) and gene expression of Nrf2 and Keap1. The results showed that curcumin could inhibited the liver damage caused by CCl_4. Curcumin at 1 μg/ml and 5 μg/ml significantly reduced the levels of GPT,GOT,LDH and AKP,increased the activities of SOD and GSH-Px,T-AOC and GSH content,suppressed the production of MDA,and up-regulated the expression of Nrf2 in the CCl_4-induced liver. In conclusion,curcumin had protective effect on liver injury induced by CCl_4. The hepatoprotective action of curcumin was probably accomplished by regulating Nrf2 expression to elevate cellular antioxidative capacity and inhibit lipid peroxidation.
