摘要
为探讨^(60)Co-γ射线对姜黄的诱变效应,本研究以姜黄种根为研究对象,采用不同剂量(60、70、80、90 Gy)的^(60)Co-γ射线进行辐射处理,分析VM_(1)和VM_(2)材料的田间表现。结果表明,姜黄种根经^(60)Co-γ射线辐射后VM_(1)生长受到明显的抑制,植株的出苗率、株高、叶长、叶宽、地上部分干重、根茎鲜重等指标均显著低于对照(CK)。VM_(2)中辐射处理的抑制作用仍然存在,但抑制效应减弱。适宜剂量辐射对姜黄VM_(2)的姜黄素类成分有促进作用,其中辐射剂量为60 Gy时,姜黄VM_(2)的姜黄素含量较CK显著提高30.7%(P<0.05);在姜黄VM_(2)中,随着辐射剂量的增加,挥发油呈先增加后降低的趋势。^(60)Co-γ射线辐射对姜黄具有诱变作用,其辐射的半致死剂量(LD50)在70~80 Gy之间。本研究结果为姜黄药材的诱变育种提供了参考依据。
To study the^(60)Co-γrays mutagenic effects on rhizome of Curcuma longa.The rhizomes of Curcuma longa were irradiated with 60,70,80 and 90 Gy of^(60)Co-γrays and the field performance of VM_(1) and VM_(2) generations were studied.In the VM_(1),the growth of Curcuma longa was significantly inhibited after irradiated by^(60)Co-γrays,and the index of seedling rate,plant height,leaf length,leaf width,dry weight of the upper part,and weight of the underground part of the plant were significantly lower than that of CK.The inhibition of irradiation in VM_(2) still exists,but the inhibition effect was weakening.Appropriate dose of irradiation had a positive effect on three curcumin ingredients of VM_(2) Curcuma longa.At the 60 Gy dose,curcumin of VM_(2) Curcuma longa was 30.7%higher than that of CK,and the difference was significant.With the increase of irradiation dose,volatile oil of VM_(2) Curcuma longa had the tendency of first increasing and then decreasing.The^(60)Co-γrays were effective to induce mutations of Curcuma longa the LD50 was 70~80 Gy.This study will provide basis for traditional Chinese medical herb breeding by irradiation.
