砂仁酚类特征成分含量测定及其特征图谱质量表征关联分析研究 被引量:10

摘要 目的:建立砂仁酚类特征指标性成分的含量测定及酚类特征图谱的质量表征关联分析方法,应用此方法和评价模式,有效精准地评价砂仁质量。方法:采用HPLC-PDA法同时测定砂仁饮片中5个特征指标性成分(原儿茶酸、香草酸、儿茶素、表儿茶素、槲皮苷)的含量,并建立砂仁饮片酚类特征图谱,基于特征图谱中特征峰的数量及其所属化学类型对11批砂仁饮片进行质的表征;基于特征图谱中特征指标性成分及黄酮类(以儿茶素表征)成分的含量和峰面积分别对11批砂仁饮片进行量的表征,并基于基准参比饮片将11批砂仁饮片质与量的表征结果进行关联性分析。结果:测定的5个特征指标性成分均具有良好的线性关系,方法学考察均符合定量测定要求;综合基于特征性成分、类型成分的含量、相对含量、峰面积及相对峰面积的质量表征,以批号5的样品为基准,批号11、1、4、2、8、10的总体含量高,批号8、6、7、10、4、3的关联性最高,整体得出:11、1、4、2、8、10、6、7等批号的饮片质量较好。结论:建立的含量测定方法简便、准确,可用于砂仁饮片特征指标性成分含量测定;构建的质量表征关联分析模式可用于分析砂仁质量整体性与应用有效性,为砂仁饮片的质量控制及评价提供可靠方法学依据。 Objective: To establish the method for quantitative determination for phenolic characteristic index components and the method correlation analysis of quantity and quality in its specific chromatogram of Fructus Amomi,thus to evaluate the quality of Fructus Amomi effectively and accurately. Methods: HPLC-PDA method was used to determine the contents of five characteristic index component simultaneously in Fructus Amomi,including protocatechuic acid,vanillic acid,catechin,epicatechin,and quercitrin; phenolic specific chromatogram was established in Fructus Amomi,on which basis,the quality of 11 batches of Fructus Amomi was represented according to the number and chemical type of characteristic peaks; the quantitative representation of the 11 batches was established on the contents and peaks area of characteristic index components and flavonoids(represented by catechin) to carry out the correlation analysis. Results:The detected five characteristic index components had good linear correlation and the results of methodological investigation were in accordance with the quantitative determination requirements; batch 5 was taken as the reference substance to evaluate the specific contents,content of the type component,relative contents,peak area,and relative peak area. It showed the overall contents of the characteristic composition of batch 11,1,4,2,8 and 10 were higher than the reference,and the highest correlation with the reference showed in batch 8,6,7,10,4 and 3. So the quality of batch 11,4,2,8,10,6 and 7 were superior to the others. Conclusion: The established method is simple and accurate,it can be used for the content determination for phenolic characteristic index components in Fructus Amomi. This evaluation mode can be applied into the analysis of overall quality of Fructus Amomi and its effectiveness,which provides a reliable basis for the comprehensive quality evaluation of Fructus Amomi.
