摘要
目的基于药物体系的发现,并运用药物体系质量评价模式全面表征远志质量并关联分析,评价远志质量。方法采用HPLC法同时测定远志中西伯利亚远志糖A5、西伯利亚远志糖A6、远志口山酮Ⅲ、3,6’-二芥子酰基蔗糖等4种酚类有效指标性成分含量,结合药典方法测定远志中萜类有效指标性成分细叶远志皂苷含量,基于药物体系质量评价模式对远志进行有效指标性成分含量及其相对比值的质量表征,并对远志5种有效指标性成分进行关联分析。使用非关联系数、非关联度、关联度等相关概念指标反映与基准饮片质量的关联性。结果样品2、3、6中有效指标性成分含量总体高于基准饮片样品4,样品9中有效指标性成分含量与样品4接近。以样品4为基准计算各样品与样品4的非关联系数,进一步得到关联度排序为:样品4(0/100%)>样品3(0.41/94.18%)>样品9(0.94/86.60%)>样品6(0.96/86.31%)>样品2(1.05/85.01%)>样品11(1.15/83.62%)>样品1(1.21/82.65%)>样品12(1.23/82.43%)>样品8(1.35/80.72%)>样品14(1.39/80.10%)>样品10(1.52/78.27%)>样品7(1.55/77.91%)>样品5(1.64/76.63%)>样品15(2.27/67.56%)>样品13(3.12/55.44%),即样品3、9、6、2与基准样品4质量关联度较高。综合质量表征及关联分析结果,样品3、6、2、9质量优良度居前,其次为样品11、1、12。结论基于药物体系质量评价模式对远志质量进行关联分析研究,可综合精准地评价远志质量优劣,为远志资源筛选、原料质量控制及应用提供了依据,同时亦可为中药质量评价提供方法学及其应用借鉴。
Objectives To establish quality-representation and perform correlation analysis method of Yuanzhi (Polygala tenuifolia Willd.) based on quality evaluation model of medicinal system to assess its quality.Methods Four phenolic compounds as effective index components,including sibricose A5,sibricose A6,polygala shantung Ⅲ,and 3,6'-disinapoy sucrose,were measured by using HPLC,while tenuifolin (terpenoids) was assessed according to China pharmacopoeia.Correlation analysis of these data was then performed.Results The content of effective index components and their content ratio as the quality index was presented based on medicinal system.Correlation analysis was performed using non-correlation coefficient,non-correlation index,and correlation index to demonstrate the correlation between samples and reference decocting pieces.The result showed the total content of effective index components in sample No.2,No.3,and No.6 was higher than that in sample No.4,whereas it was similar between sample No.9 and No.4.The correlation ranking could be produced as follows when sample No.4 was used as the reference standard to calculate non-correlation coefficient of other samples:Sample No.4(0/100%) > No.3(0.41/94.18%) > No.9(0.94/86.60%) > No.6(0.96/ 86.31%) > No.2 (1.05/85.01%) > No.11 (1.15/83.62%) > No.1 (1.21/82.65%) > No.12 (1.23/ 82.43%) > No.8(1.35/80.72%) > No.14(1.39/80.10%) > No.10(1.52/78.27%) > No.7 (1.55/ 77.91%) > No.5 (1.64/76.63%) > No.15 (2.27/67.56%) > No.13 (3.12/55.44%).Samples No.3,No.9,No.6,and No.2 had a higher correlation with the reference standard of sample No.4.Therefore,based on quality representation and correlation analysis,the quality of samples No.3,No.6,No.2,and No.9 was superior to that of samples No.11,No.1,and No.12.Conclusion Quality representation and correlation analysis of Yuanzhi based on a new quality evaluation model of medicinal system could be used to assess
