摘要
目的建立木棉花药材高效液相色谱(HPLC)指纹图谱及芒果苷含量测定方法,评价不同产地木棉花药材质量。方法采用HPLC测定15批木棉花药材的指纹图谱,结合相似度评价、热图聚类分析、主成分分析(PCA)和正交偏最小二乘法判别分析(OPLS-DA)对其指纹图谱信息进行研究,筛选出引起木棉花质量差异的主要化学成分并进行含量测定。结果建立了木棉花药材HPLC指纹图谱,共确定8个共有峰,并指认了4个共有峰。15批木棉花药材指纹图谱的相似度均大于0.990;热图分析表明不同产地木棉花药材的化学成分含量存在差异,聚类分析(CA)可将15个样品聚为2类;PCA结果显示前2个主成分的累计贡献率为83.938%,可反映原始色谱峰的大部分信息;OPLS-DA结果将样品分为3类,并筛选出2个VIP值大于1的差异性成分,分别是芒果苷和峰8;芒果苷含量与指纹图谱综合评分呈显著正相关性,均显示云南产地的木棉花药材质量更优。结论不同产地木棉花药材质量存在一定差异。本研究建立的方法可为木棉花药材的质量评价及控制提供参考。
Objective To establish HPLC fingerprints and a method for the determination of mangiferin in kapok,and evaluate the quality of kapok from different areas.Methods The fingerprints of 15 batches of kapok was determined by HPLC,and the fingerprint information was studied by combining similarity evaluation,thermogram cluster analysis,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA),and the main chemical components that caused the quality difference of kapok were screened and determined.Results The HPLC fingerprints of kapok were established.A total of 8 common peaks were marked,and 4 common peaks were identified.The similarity of fingerprints of 15 batches of kapok was greater than 0.990.The heat map showed that there were differences in the chemical composition of kapok from different sources.Cluster analysis(CA)classified the 15 samples into 2 categories.PCA results showed that the cumulative contribution rate of the first two principal components was 83.938%,which could reflect most information of the original chromatographic peaks.The samples were divided into 3 categories by OPLS-DA,and 2 different components with VIP value greater than 1 were screened out,which were mangiferin and peak 8,respectively.There was a significant positive correlation between content of mangiferin and comprehensive scores of the fingerprints.Both results showed that the quality of the kapok from Yunnan was better than that from other sources.Conclusion There were some differences in quality of kapok from different producing areas.The method established in this study can provide reference for the quality evaluation and control of kapok.
