摘要
目的建立同时测定金银花、山银花及川银花中10种成分质量分数的UPLC法,为药材质量评价提供依据。方法采用Waters BEH Phenyl色谱柱(100 mm×2.1 mm,1.7μm),以乙腈-0.1%磷酸为流动相,梯度洗脱,流速为0.3 mL/min,柱温为30℃,检测波长为240 nm,进样量为1μL;对36批忍冬科药材的10个指标成分进行测定,并采用聚类分析、主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA)进行多变量统计分析。结果金银花、山银花、川银花药材中10种成分的质量分数存在明显差异,聚类分析结果显示,三者各自聚为一类;通过主成分分析及正交偏最小二乘法,可将金银花、山银花及川银花药材进行有效区别。结论本研究建立的方法简便、准确,为不同忍冬科药材的质量控制和基原鉴别提供参考。
Objective To establish a UPLC method for simultaneous determination of the content of 10 components in Lonicerae japonicae Flos,Lonicerae Flos and Lonicerae similis Flos in order to provide the basis for the quality evaluation of medicinal materials.Methods The UPLC analysis was carried out on a Waters BEH Phenyl(100 mm×2.1 mm,1.7μm)column at a column temperature of 30℃.The mobile phase consisted of acetonitrile and 0.1%formic acid at a flow rate of 0.3 mL/min in gradient mode.The detection wavelength was 240 nm and the injection volume was 1μL.The determination of 10 components in 36 batches of different Caprifoliaceae was performed and cluster analysis,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used for the multivariate statistical analysis.Results There were significant differences in the contents of 10 components in Lonicerae japonicae Flos,Lonicerae Flos and Lonicerae similis Flos.The results of cluster analysis showed that each of Lonicerae japonicae Flos,Lonicerae Flos and Lonicerae similis Flos was classified into one category,which could be distinguished by principal component analysis and orthogonal least squares discriminant analysis.Conclusion The method is simple and accurate,which can provide a reference for the quality control and identification of different Caprifoliaceae.
