摘要
目的:采用高效液相色谱法建立鹅不食草药材的特征图谱,并同时测定7个成分的含量。方法:以绿原酸、隐绿原酸、咖啡酸、芦丁、异绿原酸B、异绿原酸A和异绿原酸C为指标,采用HSS T3 C18色谱柱(4.6 mm×250 mm,5μm),以乙腈(A)-0.08%三氟乙酸水溶液(B)为流动相,梯度洗脱(0~13 min,10%A→11%A;13~18 min,11%A→15%A;18~29 min,15%A→17%A;29~50 min,17%A),流速1.0 mL·min^(-1),检测波长为325 nm,柱温30℃;采用Chempattern化学计量学软件对结果进行处理分析。结果:建立鹅不食草特征图谱,标定7个特征峰,50 min内鹅不食草的主要色谱峰能够达到完全分离,对20批药材中7个成分进行含量测定。绿原酸、隐绿原酸、咖啡酸、芦丁、异绿原酸B、异绿原酸A和异绿原酸C质量浓度分别在1.021~102.1μg·mL^(-1)(r=0.999 9,n=6)、0.936~93.6μg·mL^(-1)(r=0.999 8,n=6)、0.952~95.2μg·mL^(-1)(r=0.999 9,n=6)、0.932~93.2μg·mL^(-1)(r=0.999 9,n=6)、0.928~92.8μg·mL^(-1)(r=0.999 9,n=6)、0.964~96.4μg·mL^(-1)(r=0.999 9,n=6)和0.927~92.7μg·mL^(-1)(r=0.999 8,n=6)范围内线性关系良好,方法的平均回收率(n=6)分别为98.6%(RSD=1.7%)、98.3%(RSD=1.7%)、98.0%(RSD=2.3%)、99.1%(RSD=1.6%)、99.1%(RSD=2.6%)、98.8%(RSD=1.8%)和97.4%(RSD=2.3%);20批鹅不食草中上述7个成分含量分别为0.009%~0.122%、0.005%~0.043%、0.005%~0.024%、0.009%~0.094%、0.005%~0.033%、0.032%~0.263%和0.012%~0.100%。结论:所建立特征图谱专属性强,结合7个主要成分含量测定能够为鹅不食草的质量控制提供科学依据。
Objective:To establish HPLC characteristic chromatogram and quantitation method of seven components for Centipedae Herba.Methods:Chlorogenic acid,cryptochlorogenic acid,caffeic acid,rutin,isochlorogenicacid B,isochlorogenic acid A and isochlorogenic acid C were determined.HPLC analysis was performed on an HSS T3 C18 column(4.6 mm×250 mm,5 μm).The mobile phase was acetonitrile-0.08% trifluoroacetic acid aqueous solution with gradient elution(0^-13 min,10%A → 11%A;13^-18 min,11%A → 15%A;18-29 min,15%A → 17%A;29-50 min,17%A).The flow rate was 1 mL·min^-1,the detection wavelength was 325 nm and the column temperature was 30 ℃.Chemometrics software Chempattern was employed to analyze the data.Results:HPLC characteristic chromatogram of Centipedae Herba was established and seven characteristic peaks were marked.Seven characteristic peaks were simultaneously determined by HPLC within 50 min.The linear ranges of chlorogenic acid,cryptochlorogenic acid,caffeic acid,rutin,isochlorogenic acid B,isochlorogenic acid C and isochlorogenic acid A were 1.021^-102.1 μg·mL^-1(r=0.999 9,n=6),0.936-93.6 μg·mL^-1(r=0.999 8,n=6),0.952-95.2 μg·mL^-1(r=0.999 9,n=6),0.932-93.2 μg·mL^-1(r=0.999 9,n=6),0.928-92.8 μg·mL^-1(r=0.999 9,n=6),0.964-96.4 μg·mL^-1(r=0.999 9,n=6) and 0.927-92.7 μg·mL^-1(r=0.999 8,n=6),respectively.The average recoveries(n=6) of seven components were 98.6%(RSD=1.7%),98.3%(RSD=1.7%),98.0%(RSD=2.3%),99.1%(RSD=1.6%),99.1%(RSD=2.6%),98.8%(RSD=1.8%) and 97.4%(RSD=2.3%),respectively.The contents of the seven components in 20 batches of Centipedae Herba were 0.009%-0.122%,0.005%-0.043%,0.005%-0.024%,0.009%-0.094%,0.005%-0.033%,0.032%-0.263% and 0.012%-0.100%,respectively.Conclusion:The proposed specific HPLC characteristic chromatogram and quantitation method of seven components for Centipedae Herba offered reference for quality control of the crude drug.
