摘要
目的:探讨NO、Gal-9在宫颈癌血清中的表达及二者与宫颈癌细胞凋亡的关系,并分析血清NO、Gal-9在宫颈癌中表达的相关性。方法:利用硝酸还原酶法分别检测宫颈鳞状细胞癌患者、CIN Ⅱ-Ⅲ患者及对照组患者血清中NO的含量;选用酶联免疫吸附试验(ELISA)法检测各组患者血清中Gal-9蛋白的表达水平;原位末端标记(TUNEL)法检测相应各组患者宫颈细胞的凋亡情况,计算凋亡指数(apoptotic index,AI)。并对三项指标进行相关性分析。结果:CSCC组与对照组比较:血清NO及Gal-9水平显著升高(P<0.05);宫颈细胞AI值明显升高(P<0.05)。CIN Ⅱ-Ⅲ组与对照组比较:血清NO及Gal-9水平明显升高(P<0.05);宫颈细胞AI值显著升高(P<0.05)。CSCC组与CIN Ⅱ-Ⅲ组比较:血清NO及Gal-9水平升高(P<0.05);宫颈细胞AI值升高(P<0.05)。血清NO与Gal-9之间呈显著正相关(P<0.01);同时,血清NO及Gal-9均与宫颈细胞AI值呈正相关(P<0.01)。结论:血清中NO、Gal-9的过度表达参与了HRHPV相关的宫颈鳞状细胞癌的发生发展及细胞凋亡;宫颈癌血清中NO和Gal-9具有正相关性。
Objective:To investigate the expression of NO and Gal-9 in the serum of cervical cancer and the relationship between the two and the apoptosis of cervical cancer cells,and to analyze the correlation between the expression of serum NO and Gal-9 in cervical cancer.Methods:The content of NO in serum of patients with cervical squamous cell carcinoma,CIN II-III and the control group was detected by nitric acid reductase method.Gal-9 expressionin serum was detected by enzyme-linked immunosorbent assay (ELISA).Apoptosis of cervical cells was detected by transferase-mediated deoxyuridine triphosphate-biotin nick end labeling(TUNEL),and the apoptosis index (AI) was calculated.Then the correlation of the three indicators was analysed.Results:Comparison between CSCC group and control group:Serum NO and Gal-9 levels were significantly elevated (P〈0.05),the AI of cervical cells increased significantly (P〈0.05).Comparison between CIN II-III group and control group:Serum NO and Gal-9 levels were significantly elevated (P〈0.05),the AI of cervical cells increased significantly (P〈0.05).Comparison between CSCC group and CIN II-III group:Serum NO and Gal-9 levels were elevated (P〈0.05),the AI of cervical cells increased significantly(P〈0.05).There was significant positive correlation between serum NO and Gal-9 (P〈0.01).Serum NO and Gal-9 were positively correlated with the AI of cervical cells(P〈0.01).Conclusion:Overexpression of NO and Gal-9 in serum is involved in the occurrence,development and apoptosis of the HR-HPV associated cervical squamous cell carcinoma.The serum NO and Gal-9 have positive correlation in cervical cancer.
