摘要 目的 :探讨新的凋亡相关基因 Survivin蛋白在宫颈癌中的表达及其与肿瘤细胞凋亡的关系。方法 :采用免疫组织化学 S- P法和 DNA原位末端标记 TUNEL法分别测定45例宫颈癌组织及 1 0例正常宫颈组织中 Survivin蛋白的表达和细胞凋亡。结果 :Survivin蛋白在宫颈癌组织中的阳性表达率为 68.89% ,与病理分级、临床分期和淋巴结转移密切相关 ( P<0 .0 5 ) ,而正常宫颈组织中无阳性表达 ;宫颈癌组织及正常宫颈组织中细胞凋亡指数( AI)分别为 7.5 5± 2 .35、5 .64± 1 .81 ,其差异有显著意义 ( P<0 .0 5 ) ;Survivin蛋白的表达与细胞凋亡指数呈负相关 ( r=- 0 .5 4 1 ,P<0 .0 1 )。结论 :Survivin蛋白的异常表达而引起的细胞凋亡抑制或逃避 ,在宫颈癌的发生、发展中起一定的作用 ;联合检测 Survivin蛋白和凋亡指数 ,有助于对肿瘤细胞的分化程度作出正确评价 。 Ojective: To investigate the expression of survivin proteins in tissues of cervical carcinomas(CC) and its correlation with apoptosis of cancer cells. Methods: Expression of survivin proteins and apoptotic index(AI) were detected by immunohistochemical and terminal deoxynucleotidyl transterase mediated dUTP biotin nick end labeling(TUNEL) technique in 45 cases of cervical carcinoma tissues and 10 cases of normal cervical tissues.Results: Survivin proteins were expressed in 31 of 45 cases of CC(68.89%),the positive rate of survivin was strongly associated with pathological,clinical stage and lymph metastasis ( P <0.05).In contrast,normal cervical tissues did not express survivin. Survivin proteins expression was negatively correlated with AI in CC( r =-0.541, P <0.01). Conclusion: Apoptosis inhibition or escapement induced by obnavmal expression of survivin may participate in the onset and development of CC. Detection of survivin protein and AI in CC may be helped to evaluate the degree of cell deferentiation and to decide therapeatic strategies and to estimate prognosis.
