摘要
目的探讨姜黄素对糖尿病心肌病大鼠心肌保护作用以及对神经调节蛋白-1(neuregulin-1,NRG-1)/Erb B2信号通路的影响。方法将80只Wistar大鼠随机分为正常组(CN组)、模型组(M组)、姜黄素高剂量组(HC组)、姜黄素低剂量组(LC组)和人类重组NRG-1药物组(AD组),检测各组大鼠血清中生化指标以及心肌功能变化情况,蛋白免疫印迹法(western blotting)检测NRG-1/Erb B2信号通路中蛋白表达情况,实时荧光定量聚合酶链反应(RT-PCR)检测Bax、Bcl-2基因表达情况。结果 HC组空腹血糖(fasting blood-glucose,FPG)、三酰甘油(triglycerides,TG)、肌酸激酶同工酶-MB(creatine kinase-MB,CK-MB)、左心室舒张末内径(left ventricular end diastolic dimension,LVEDD)、左心室收缩末内径(left ventricular end systolic dimension,LVESD)及左心室后壁厚度(left ventricular posterior wall thickness,LVPWT)均显著低于M组,差异有统计学意义(P<0.05或P<0.01);而胰岛素(insulin,INS)、左心室射血分数(left ventricular ejection fraction,LVEF)、左心室短轴缩短率(left ventricular fractional shortening,LVFS)及心率(heart rate,HR)显著高于M组,差异有统计学意义(P<0.01)。苏木素-伊红染色和Masson染色显示,M组心肌组织中细胞排列紊乱,心肌纤维肥大、疏松,可见纤维空泡样变化,而AD组、HC组心肌细胞排列整齐,心肌间质未见异常改变。TUNEL检测结果显示,M组凋亡指数(apoptotic index,AI)显著高于CN组,差异有统计学意义(P<0.05);而AD组、LC组及HC组AI显著低于M组,差异有统计学意义(P<0.05)。Western blotting结果显示,LC组及HC组NRG-1、p-Erb B2蛋白相对表达量均显著高于M组,差异有统计学意义(P<0.05);而转化生长因子β1(TGF-β1)蛋白相对表达量显著低于M组,差异有统计学意义(P<0.05)。逆转录-聚合酶链反应检测结果显示,HC组Bax基因相对表达量显著高于M组,而Bcl-2基因相对表达量显著低于M组,差异均有统计学意义(P<0.05)。结论姜黄�
Objectives To study the protective effect of curcumin on the myocardium of diabetic cardiomyopathy rats and its effect on the neuregulin-1(NRG-1)/ErbB2 signaling pathway. Methods Totally 80 Wistar rats were randomly divided into normal group(CN group),model group(M group),high dose of curcumin group(HC group),low dose of curcumin group(LC group)and recombinant human NRG-1 drug group(AD group). Biochemical indexes in serum and changes of myocardial function were measured. Protein expression in NRG-1/ErbB2 signal pathway was detected by western blotting. Gene expressions of Bax and Bcl-2 were detected by quantitative real-time polymerase chain reac?tio (QRT-PCR). Results In HC group,fasting blood-glucose (FPG),triglycerides(TG),creatine kinase-MB (CK-MB),left ventricular end diastolic dimension(LVEDD),left ventricular end systolic dimension(LVESD)and left ventricular posterior wall thickness(LVPWT)were significantly lower than those in M group(P〈0.05 or P〈0.01);insulin(INS),left ventricular ejection fraction(LVEF),left ventricular fractional shortening(LVFS)and heart rate(HR)were significantly higher than those of M group(P〈0.01). TUNEL assay showed that apoptotic index(AI)in Mgroup was significantly higher than that in CN group(P〈0.05). AI in AD group,LC group and HC group wassignificantly lower than that of M group(P〈0.05).
