摘要
目的:探讨姜黄素对糖尿病心肌病大鼠心肌NF-κB的影响和机制。方法:采用SD大鼠尾静脉注射45 mg/kg链脲佐菌素建立实验性糖尿病模型,30只成模大鼠随机分为糖尿病心肌病组(DCM组)、姜黄素治疗组[Curcumin-L组,15 mg/kg·d;Curcumin-H组,30 mg(kg·d)],正常对照组(NC组)10只。用药处理8周后检测各组大鼠体质量、血糖、心脏重量指数,masson染色观察各组心肌组织变化并计算心肌胶原容积分数(CVF),Real-time PCR测定miRNA-146a水平,Western-blot测定NF-κB。结果:与DCM组比较,姜黄素治疗组大鼠血糖水平无明显变化,但心脏重量指数明显降低。Masson染色DCM组心肌细胞排列紊乱,肌纤维间纤维组织增生明显,姜黄素治疗组心肌细胞排列较DCM组规则,纤维组织增生明显减少。与DCM组比较,姜黄素组CVF水平明显降低(P<0.05),miRNA-146a表达上调(P<0.05),NF-κB表达下调(P<0.05)。结论:姜黄素通过抑制糖尿病大鼠心肌NF-κB的表达而对糖尿病大鼠心肌有保护作用,其机制与miRNA-146a有关。
Objective : To investigate the effect and mechanism of curcumin on NF - KB in myocardium of diabetic cardiomy0p- athy rats. Methods:Rats were randomly divided into diabetic cardiomyopathy group (DCM) and curcumin group [ curcumin low dose group, 15 mg/( kg . d) ;curcumin high dose group,30 mg/( kg . d) ) ]. Normal group was 10 rats. Eight weeks later, body mass, blood glucose and cardiac mass index were detected. Masson method was used to observe cardiac muscle tissue and collagen volume fraction (CVF) was calculated. Real - time PCR was applied to detect miRNA - 146a. western - blot was for detecting NF - KB. Results : Compared with DCM group, blood glucose was not obviously changed, but cardiac mass index was decreased of curcumin group. Myocardial cell was disorganized and fibrosis was hyperplastic by masson method of DCM group. Myocardial cell was organized and fibrosis was not hyperplastic of curcumin group. Compared with DCM group, CVF was decreased, miRNA - 146a was increased and NF - KB was decreased ( P 〈 0.05 ) of curcumin group. Conclusion : Curcumin protected diabetic rats my ocardium through inhibiting NF - KB expression. The mechanism was related to miRNA - 146a.
