不同激素对杂交魔芋愈伤诱导及植株再生的影响 被引量:4

Effect of Different Hormones on Callus Induction and Plant Regeneration of Hybrid Amorphophallus konjac

摘要 以杂交魔芋球茎为外植体,以添加不同种类和浓度的生长素及细胞分裂素的MS为培养基,研究不同浓度激素配比对魔芋愈伤组织诱导形成、增殖、分化以及植株再生等的影响。结果表明:6-BA浓度在3.0 mg/L时,对魔芋愈伤组织的诱导率最高,达到90%;6-BA与NAA配合使用时,有利于提高愈伤组织分化率,6-BA浓度在0.5~1.0 mg/L、NAA浓度在0.1~0.5 mg/L时,成苗率均较高,其中6-BA浓度在1.0 mg/L、NAA浓度在0.1 mg/L时,愈伤分化率最高,达到84.4%,成苗率达到253%;NAA浓度在0.2~0.5 mg/L时,生根率最高,达到247%。综合以上不同浓度及激素配比的作用效果,认为适合杂交魔芋麻杆球茎植株再生的最适愈伤诱导培养基是MS+6-BA 3.0 mg/L;最适愈伤分化及芽分化培养基是MS+6-BA 1.0 mg/L+NAA 0.1 mg/L;最适生根培养基是1/2 MS+NAA 0.2~0.5 mg/L。 Corms of hybrid A morphophallus konjac were used as explants and cultured in the MS medium at presence of different concentrations of hormones (auxin and cytokinin) to observe the effect of different combinations of hormones on callus formation, proliferation, differentiation and plantlet regeneration of the hybrid konjac. The results showed that the hybrid konjac had a calli inducing rate of 90%, the highest, when cultured in the medium supplemented with 6-BA at a concentration of 3.0 mg/L, and the shoot formation rate was higher when the concentrations of 6-BA and NAA were at the range of 0.5 ~ 1.0 mg/L and 0.1 ~0.5 mg/L, respectively; when the concentrations of 6-BA and NAA were 1.0 mg/L and 0. I mg/L, respectively, the callus differentiation rate was 84.4%, the highest, and the shoot formation rate was upto 253%; when the NAA concentration was 0.2~0.5 mg/L, the rate of rooting was the highest (247%). The results showed that MS+6-BA 3.0 mg/L was the best medium for callus induction, that MS+6-BA 1.0 mg/L+NAA 0.1 mg/L was the best medium for callus and shoot differentiation, and that 1/2 MS+NAA 0.2 ~0.5 mg/L was the best medium for rooting.
