摘要
为解决杂交魔芋(Amorphophallus konjac)组织培养在愈伤组织分化成芽这一阶段出现的分化时间长、分化效果差、分化芽数少等问题,设计不同激素不同浓度间的正交试验,以期找到能促进愈伤组织快速有效分化成芽的最优方案。结果显示:对促进愈伤组织快速有效分化成芽的最优组合为A3B2C2,即6-BA 3.0mg/L+NAA 0.5mg/L+KT 0.5mg/L,对促进生成最多芽数的最优组合为A3B1C2,即6-BA3.0mg/L+NAA 0.2mg/L+KT 0.5mg/L。综合来看,既能促进愈伤组织快速有效分化,又能促进其生成更多的芽的最有效组合为A3B2C2,即6-BA 3.0mg/L+NAA 0.5mg/L+KT 0.5mg/L。
In order to deal with the problems such as the long time of differentiation,poor differentiation effect and low differentiation of buds at the stage of the callus differentiate to buds,orthogonal test with different concentrations of different hormones was designed to find the optimal solution for the rapid and effective differentiation of the callus.The results showed that A3B2C2(6-BA3.0 mg/L+NAA 0.5 mg/L+KT 0.5 mg/L)was the optimal combination for the rapid and effective differentiation of the callus,and the optimal combination for the maximum number of buds was A3B1C2(3.0 mg/L + NAA 0.2 mg/L+ KT 0.5 mg/L).Generally,the most effective combination was A3B2C2(6-BA 3.0 mg/L +NAA 0.5 mg/L+KT 0.5 mg/L),which could promote rapid and effective differentiation of callus and promote the formation of more buds.
