高效液相色谱法测定丹参中9种非法添加的合成色素 被引量:5

摘要 目的建立高效液相色谱法同时测定丹参中柠檬黄、日落黄、酸性橙等9种黄色合成色素的方法。方法样品经无水乙醇-水(7∶3,v/v)超声提取,以Tech Mate ST-C18柱(4.6×250 mm,5μm)分离,以甲醇0.05 mol/L的乙酸铵为流动相进行梯度洗脱,流速1.0 mL/min,柱温35℃,检测波长为405 nm(酸性黄17、亮黄)、430 nm(柠檬黄、金胺O)、485 nm(日落黄、酸性橙10、碱性橙21、金橙Ⅱ、碱性橙22)。结果本法建立色谱条件下九种合成色素专属性较好,无干扰峰。质量浓度在1.00~50.0 mg/L范围内呈良好线性关系,r=0.999 9,3个水平下平均加标回收率范围99.24%~101.73%,RSD<2%。结论该方法具有简单、快速、灵敏度高、稳定性好、回收率高、适用范围广的特点,因此可用于中药材丹参中9种合成色素的快速检测。 Objective To develop a high performance liquid chromatography with multiple wavelength method for simultaneous determination of 9 kinds of yellow synthetic pigment(Tartrazize,Sunset yellow,Acid orange,etc.)in salvia miltiorrhiza.MethodsThe samples were extracted with ultrasonic from anhydrous ethanol: water (7∶3,v/v) and separated on TechMate ST-C18 column (4.6&#215;250 mm,5 μm),by a gradient elution using methano (A) and 0.05 moL/L ammonium acetate(B) as mobile phase at flow rate of 1.0 mL/min, and the column temperature was 35 ℃.The wavelengths for detection were 405 nm(Acid yellow 17,Brillant yellow),430 nm(Tartrazize,Auramine O),485 nm(Sunset yellow,Acid orange 10,Basic orange 21, aurantia Ⅱand Basic orange 22).Results Under such chromatographic conditions,the specificity of the nine kinds of synthetic pigments was good,and there was no interference peaks.The calibration curves of 9 kinds of synthetic pigments showed good linear relationship in the range of 1.00~50.0 mg/L,r=0.999 9.The mean standard recovery rates ranged from 99.24% to 101.73%,RSD 〈2%.ConclusionThe method is simple,rapid,sensitive,stable,with high recovery rate and wide range of application.Therefore,it can be used for simultaneous determination of 9 kinds of synthetic pigments in salvia miltiorrhiza.
