胭脂红对3T3细胞DNA损伤的SCGE检测 被引量:12

摘要 目的研究人工合成偶氮类色素胭脂红对体外培养小鼠胚胎成纤维细胞株(NIH3T3)DNA的损伤作用。方法单层培养的3T3细胞中加入不同浓度胭脂红培养4 h,用碱性单细胞凝胶电泳技术(single cell gel elec-trophoresis,SCGE)检测3T3细胞DNA的单链断裂(single-strand breaks,SSBs)情况。结果各染毒组彗星细胞率、彗星细胞的尾长、尾部DNA含量及尾矩与胭脂红浓度正相关,呈剂量-反应关系。结论胭脂红对体外培养3T3细胞DNA具有损伤作用,浓度越大,损伤越严重。 Objective Study DNA damage of mouse NIH3T3 cells induced by ponceau 4R, an artificial synthetic azo pigment, in vitro. Methods The 3T3 cells were exposed to ponceau 4R with different concentrations for 4 hours, and the DNA single-strand breaks(SSBs) of 3T3 cells were determined by alkaline single cell gel electrophoresis(SCGE). Results The proportion of comet 3T3 cells, the tail length, DNA contents in tail and tail moment of comet cells had positive correlations with the concentration of ponceau 4R, and the dose-response relationship was shown. Conclusion DNA of 3T3 cells in vitro can be damaged by a certain amount of ponceau 4R, and more damages were found with the increase of ponceau 4R concentration.
