高效液相色谱法同时测定丹参药材和饮片中苋菜红、胭脂红、金胺O含量 被引量:3

Simultaneous Determination of Amaranth,Carmine and Auramine O in Salviae Miltiorrhizae Radix et Rhizoma and Its Pieces by HPLC

摘要 目的建立同时测定丹参药材和饮片中苋菜红、胭脂红、金胺O含量的高效液相色谱法。方法色谱柱为中谱红RD-C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.05 mol/L乙酸铵溶液(梯度洗脱),流速为1.0 mL/min,柱温为35℃,检测波长为512 nm(苋菜红和胭脂红)、432 nm(金胺O),进样量为10μL。结果苋菜红、胭脂红、金胺O质量浓度均在1~50μg/mL范围内与峰面积线性关系良好(r≥0.9993,n=5);检测限分别为0.10,0.10,0.03 mg/kg,定量限分别为0.33,0.33,0.10 mg/kg;精密度和稳定性试验的RSD均小于2.0%;平均加样回收率分别为95.93%,94.70%,96.33%,RSD分别为1.03%,1.21%,1.17%(n=6)。2批丹参药材中检出金胺O,含量分别为0.16 mg/kg和0.21 mg/kg;21批丹参饮片中均未检出上述3种色素,或检出量低于定量限。结论该方法操作简便、灵敏度好、准确度高,可用于丹参药材和饮片中苋菜红、胭脂红、金胺O的含量测定。 Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of amaranth,carmine and auramine O in Salviae Miltiorrhizae Radix et Rhizoma and its pieces.Methods The chromatographic column was Zhongpu Hong RD-C18column(250 mm×4.6 mm,5μm),the mobile phase was methanol-0.05 mol/L ammonium acetate solution(gradient elution),the flow rate was 1.0 mL/min,the column temperature was 35℃,the detection wavelength was512 nm(amaranth,carmine)and 432 nm(auramine O),and the injection volume was 10μL.Results The linear range of amaranth,carmine and auramine O was 1-50μg/mL(r≥0.9993,n=5).The limits of detection(LOD)of amaranth,carmine and auramine O were 0.10,0.10 and 0.03 mg/kg respectively,and the limits of quantification(LOQ)of the above three pigments were 0.33,0.33 and 0.10 mg/kg respectively.The RSDs of precision and stability tests were lower than 2.0%.The average recoveries of amaranth,carmine and auramine O were 95.93%,94.70%and 96.33%with RSDs of 1.03%,1.21%and1.17%(n=6).Auramine O was detected in the two batches of Salviae Miltiorrhizae Radix et Rhizoma,with the contents of 0.16 mg/kg and 0.21 mg/kg respectively.Amaranth,carmine and auramine O were not detected in the 21 batches of Salviae Miltiorrhizae Radix et Rhizoma Pieces,or the LOD of the above three pigments were lower than their LOQ.Conclusion The method is simple,sensitive and accurate,which can be used for the determination of amaranth,carmine and auramine O in Salviae Miltiorrhizae Radix et Rhizoma and its pieces.
