摘要
目的:建立丹参药材HPLC指纹图谱,研究丹参提取物指纹图谱与抗氧化活性之间的谱效关系。方法:采用HPLC法建立丹参的指纹图谱;使用“中药色谱指纹图谱相似度评价系统(2012.130723版)”进行相似度评价;分别采用ABTS法及FRAP法检测总抗氧化活性;以总抗氧化活性为药效指标,采用偏最小二乘回归法分析其谱效关系。结果:丹参醇提物指纹图谱中共有峰数为14个,其中x8为内标峰,未计算在内,其余13个共有峰中,7个与药效呈正相关,6个呈负相关,x_(1)、x_(3)、x_(5)、x_(6)为主要药效峰。结论:丹参提取物具有较强的抗氧化作用,其抗氧化活性是多种成分共同作用的结果。
Objective:To establish high performance liquid chromatography(HPLC)fingerprint of Danshen(丹参)and study the relationship between the HPLC fingerprint and antioxidant activity.Methods:The HPLC fingerprint of Danshen(丹参)was established and the SOP of similarity evaluation system for chromatographic fingerprint of TCM(Version 2012.130723)was employed to evaluate the similarity.Total antioxidant activity was detected by ABTS and FRAP,respectively.With antioxidant activity as pharmacodynamic index,the relationship between the fingerprint and the efficacy was analyzed by partial least squares regression.Results:There were 14 common peaks in the HPLC fingerprint of ethanol extract of Danshen(丹参),among which x8 was the peak for the internal standard and was not included in the calculation.Among the rest 13 common peaks,7 were in positive correlation and 6 peaks were in negative correlation with the efficacy.x_(1),x_(3),x_(5),x_(6)were the main peaks for the antioxidant activity.Conclusion:Danshen(丹参)has strong antioxidant activity thanks to the multiple components.
