摘要
从T细胞亚群变化的角度探讨补肾活血汤治疗免疫性卵巢早衰的免疫调控机制。对B6AF1小鼠进行皮下注射ZP3多肽,建立自身免疫性卵巢早衰小鼠模型,用补肾活血汤进行灌胃治疗,实验结束后,计算脏器指数,对卵巢进行H&E染色和ZP3免疫荧光染色来检测卵巢组织形态变化和透明带抗体分布,流式细胞术检测脾脏中CD4^+T细胞、CD8^+T细胞、CD4^+CD25^+Treg细胞和CD4^+IL-17^+Th17细胞比例。结果显示补肾活血汤能够提高免疫性卵巢早衰小鼠的卵巢指数,减少卵巢组织中炎症细胞浸润,增加卵泡的数量,减少卵巢组织中透明带抗体的产生;补肾活血汤能够下调脾脏中CD8^+T细胞和Th17细胞的比例,上调脾脏中CD4^+T细胞和Treg细胞的比例。表明补肾活血汤能够通过调节T细胞亚群的平衡,减轻炎症反应的程度,从而改善免疫性卵巢早衰。
To explore the immunoprotective effects and the underlying mechanisms of BSHXF on POF from Treg cell, the mouse model of immunological premature ovarian failure was established by injecting ZP3 polypeptide subcutaneously to the B6AF1 mice. The B6AF1 mice were grouped and administrated by Bushenhuoxue Soup. All mice were sacrificed and organ index was calculated. H&E staining and immunofluorescence staining were used to detect the ovarian tissue morphology and ZP3 expression, flow cytometry was used to detect the ratio change of spleen CD4+T cells, CD8 +T cells, CD4+ CD25+ Treg cells and CD4 + IL-17+ Th17 cells. By the test, it is found that Bushenhuoxue Soup could increase ovarian index, reduce ovarian tissue lymphocyte infiltration, increase the number of follicles,and reduce the production of ovarian tissue zona pellucida antibodies. Bushenhuoxue Soup could decrease the proportion of spleen CD8+ T cells and Th17 cells and increase the ratio of CD4+ T cells and Treg cells. So Bushenhuoxue Soup can alleviate inflammation and improve immunological premature ovarian failure via regulating the balance of T cell subsets.
