摘要
目的:建立柴胡桂姜颗粒中黄芩苷的含量测定方法.方法:采用高效液相色谱法,色谱柱为Agilent Extend ODS反相柱(4.6 mm×250 mm,5μm),流动相为甲醇-水-磷酸(45:55:0.2),流速为1.0 mL/min,检测波长为280 nm.结果:黄芩苷在0.025-0.100 mg/mL范围内进样量与峰面积呈良好的线性关系,r=0.9998;精密度、稳定性和重复性均良好;平均加样回收率为99.43%,RSD=1.61%.结论:本法简单、快速、专属性强、准确度高,可作为柴胡桂姜颗粒中黄芩苷的含量测定方法.
Objective: To develop a method for determination of baicalin content in chaihu guijiang granules. Methods: HPLC was adopted using Agilent Extend ODS reversed-phase column (4.6 mm × 250 mm, 5 μm),serving methanol-water-phosphoric acid (45 : 55 : 0.2)as mobile phase at a flow rate of 1.0 mL/min.The wavelength for detection was 280 nm. Results: The baicalin at a concentration of 0.025 - 0.100 mg/mL showed good linear relationship to the peak area (r = 0.999 8 ).The method showed high precision, stability and reproducibility.The average recovery rate was 99.43% ,with a RSD of 1.61%. Conclusion: The method was simple,rapid,and showed good specificity and accuracy, which might be used for determination of baicalin content in chaihu guijiang granules.
