摘要 目的 建立一种RP-HPLC测定知母及其总黄酮提物、片剂中芒果苷和新芒果苷含量的方法。方法 采用色谱柱为C18-ODS反相柱,Hypersil填料(200mm×4.6mm,5μm),流动相为乙腈-0.05 mol·L-1 NaH2PO4(pH3.20)(10:90),检测波长317nm。结果 芒果苷和新芒果苷分别在2.0-40.5,3.0-59.2μg·mL-1的浓度内呈良好的线性关系(r=0.999 7,0.999 5)。回收率分别为96.4%(RSD=1.79%,n=5),97.1%(RSD=2.14%,n=5)。结论本法简便,准确,灵敏度高,重现性好,可用于该药材及其制剂原料中芒果苷和新芒果苷的含量测定。 OBJECTIVE: To establish a HPLC assay for the determination of mangiferin and neomangiferin in Rhizoma Anemarrhenal as well as its total flavones. METHODS: The determination was performed on a C18-ODS column (200 mm x 4.6 mm, 5 μm), with the mobile phase of acetonitrile-0.05 mol&middotL -1 NaH2PO4 (adjusting pH to 3.20) (10:90). The detection wavelength was at 317 nm. RESULTS: Both of mangiferin and neomaniferin showed good linearity over the ranges of 2.0 ∼ 40.5 μg.mL-1 (r = 0.999 7)and 3.0 ∼ 59.2 μg.mL-1 (r = 0.999 7), respectively. The recoveries were 96.4% (RSD = 1.79%, n = 5) for mangiferin and 97.1% (RSD = 2.14%, n = 5) for neomangiferin. CONCLUSION: This method was easy, rapid, accurate, and sensitive. It will be employed for determining mangiferin and neomangiferin in herbs and tablets.
