肉苁蓉颗粒剂对帕金森病大鼠模型黑质纹状体多巴胺能神经元的保护作用研究 被引量:19

摘要 目的:探讨肉苁蓉颗粒剂对帕金森病大鼠模型黑质纹状体多巴胺能神经元的保护作用。方法:采用6-羟基多巴胺单侧双靶点颅内定向注射法制备帕金森病大鼠模型,3周后据阿朴吗啡诱导的不自主运动,筛选造模成功大鼠,随机分为空白组,模型组,假手术组,肉苁蓉颗粒剂低剂量组(1 g·kg-1·d-1)和肉苁蓉颗粒剂高剂量组(3 g·kg-1·d-1),灌胃相应药物共4周。记录阿朴吗啡诱导的大鼠向健侧旋转的圈数;采用免疫组化研究观察黑质内酪氨酸羟化酶(TH)的表达;和免疫印迹法检测(Western-blot)观察纹状体内α-突出核蛋白(α-synuclein)、酪氨酸羟化酶(TH)、多巴胺转运蛋白(DAT)的表达;高效液相色谱-质谱联用(HPLC-MS/MS)法检测定大鼠纹状体多巴胺(DA)、3,4-二羟基苯乙酸(DOPAC)、高香草酸(HVA)的含量。结果:用药4周后,肉苁蓉组向健侧旋转的圈数与模型组比较明显减少(P<0.05),黑质内TH阳性细胞较模型组明显增多,纹状体内α-synuclein表达下调、TH、DAT表达上调。纹状体DA、DOPAC含量明显升高(P<0.05或P<0.01)。高剂量组效果优于低剂量组。结论:肉苁蓉颗粒对帕金森病大鼠模型黑质和纹状体的多巴胺能神经元具有神经保护作用。 Objective: To study the Cistanche Granule's nigrostriatal dopaminergic protection on rat model of Parkinson's disease.Methods: The rat model of Parkinson's disease was prepared by using the 6-OHDA unilateral double target.The successful model rats were randomly divided into control group,model group,sham group,Cistanche low dose group(1 g·kg^-1·d^-1) and Cistanche Granules high dose group(3 g·kg^-1d-1).All groups were administered with corresponding medicine by intragastric administration(i.g) for 28 days.The apomorphine-induced contralateral rotation cycles were recorded.Immunohistochemically study was used to study tyrosine hydroxylase(TH) expression in the substantia nigra.And Western blotting was used to observe the expressions of α-synuclein,TH,dopamine transporter(DAT) in the striatum.High performance liquid chromatography-mass spectrometry(HPLC-MS/MS) was used to detect striatal dopamine(DA),3,4-dihydroxyphenyl acetic acid(DOPAC) and homovanillic acid(HVA) levels.Results: After 4 weeks of medication,Cistanche group's contralateral rotation cycles was significantly decreased compared with the model group's(P〈0.05).TH positive cells in the substantia nigra were significantly increased compared with those of the model group.The expression of α-synuclein was lowered while DAT and TH up-regulated in the striatum.The contents of DA and DOPAC were significantly increased(P〈0.05 or P〈0.01).The effect of high dose group was better than that of low dose group.Conclusion Cistanche Granule has nigrostriatal dopaminergic protection on rat model of Parkinson's disease.
