松果菊苷对帕金森病模型大鼠纹状体内质网应激相关凋亡蛋白ASK1的影响 被引量:4

Effect of Echinacoside on Striatum Endoplasmic Reticulum Stress Related Apoptosis Protein ASK1 in Parkinson’s Disease Model Rats

摘要 目的:观察松果菊苷对帕金森病模型大鼠内质网应激相关凋亡蛋白ASK1、caspase-12的影响,探讨松果菊苷治疗帕金森病的作用机制。方法:建立帕金森病雄性SD大鼠模型16只,随机分为模型组及治疗组,并设立正常组和溶剂对照组,每组各8只,造模结束后治疗组给予松果菊苷(20mg/kg)灌胃,其余组蒸馏水灌胃。连续14天。各组给药前1天、给药第14天分别进行行为学步幅试验;药物干预结束后,麻醉下处死大鼠取纹状体,Western Blot检测ASK1、caspase-12蛋白的表达。结果:在步幅试验中,给药前1天模型组及治疗组步幅距离较正常组减少(P<0.05),给药14天后治疗组步幅距离较模型组有所延长。与正常组比较模型组及松果菊苷组中ASK1、caspase-12蛋白表达增加(P<0.05),与模型组比较松果菊苷组ASK1、caspase-12蛋白表达减少(P<0.05)。结论:松果菊苷能改善帕金森病模型大鼠部分行为障碍,可减少纹状体ASK1、caspase-12蛋白表达,并对纹状体神经元起到保护作用。 Objective:To observe the effect of echinacoside on the endoplasmic reticulum stress-related apoptosis proteins ASK1 and caspase-12 in Parkinson’s disease model rats and to explore the mechanism of action of echinacoside in the treatment of Parkinson’s disease. Method: Sixteen male SD rats with Parkinson’s disease were randomly divided into model group and treatment group. Normal group and solvent control group were established, with 8 rats in each group. After treatment, the treatment group were given echinacoside 20 mg./kg in gavage while the other groups were given distilled water in gavage. They were with continuous administration for 14 days. The behavioral stride test was performed 1 day before the administration and on the 14 th day after the administration. After the drug intervention, the rats were sacrificed under anesthesia and the striatum was taken. The expression of ASK1 and caspase-12 protein was detected by Western Blot. Result: In the stride test, the stride distance between the model group and the treatment group was lower than that of the normal group 1 day before the administration(P<0.05). After 14 days of administration, the stride distance of the treatment group was longer than that of the model group. Compared with that of the normal group, the expression of ASK1 and caspase-12 protein in the model group and the echinacoside group increased(P<0.05). Compared with that of the model group, the expression of ASK1 and caspase-12 protein in the echinacoside group decreased(P<0.05). Conclusion: Echinacoside could partially improve the behavioral disorder of rats with Parkinson’s disease, reduce the expression of ASK1 and caspase-12 protein in striatum and protect the striatum neurons.
