银杏平颤方及其拆方对MPTP模型小鼠脑内DA及其代谢产物含量的影响 被引量:10

摘要 目的 :观察银杏平颤方及其拆方对帕金森病 (PD)模型小鼠行为学和前脑纹状体多巴胺及其代谢产物含量的影响。方法 :采用爬杆法测量小鼠行为学改变 ,应用高压液相电化学法测定前脑多巴胺(DA)、高香草酸 (HVA)及二羟基苯乙酸 (DOPAC)。结果 :PD小鼠出现爬杆能力下降、时间延长、自发活动次数减少 ,中药全方组和拆方各组小鼠行为学异常均可不同程度改善 (P均 <0 0 5 ) ;中药各组的DA含量较正常组低 (P均 <0 0 0 1) ,DA HVA、DA DOPAC比值均明显降低 (P <0 0 1或P <0 0 0 1) ;拆方组中化瘀通络 (H)组HVA、DOPAC的含量明显增加 ,但DA的含量与模型组相比差异不显著 (P >0 0 5 )。结论 :银杏平颤方拆方组中化瘀通络 (H)组可部分改善PD模型小鼠的行为异常改变 ,并能调节前脑纹状体DA与其代谢产物含量及DA HVA、DA DOPAC比值。 To investigate the effect and its mechanism of Chinese herb recipe in the treatment of Parkinson's disease (PD),we observe the effect of Yinxingpingchan Recipe and its three similar prescriptions on the abnormal behavior,dopamine(DA)metabolism of model C57/BL mice,which was established by 1-methyl-4-phenyl1,2,3,6-tetrahydropyridine(MPTP).According to high performance liquid chromatography (HPLC), Chinese medicine groups treated mice showed dominant decrease in content of DA and the ratio of DA/DOPAC,DA/HVA( P <0.001,Dome P <0.01)compare with Normal group;Chinese medicine group Huayutongluo(H)treated mice showed dominant increase in content of DA,DOPAC( P <0.05),HVA( P <0.01)compare to Model group(M).It maybe show that Huayutongluo(H) prescription of Yinxingpingchan Recipe and Yinxingpingchan Recipe all could improve on the abnormal behavior by eliciting an increase of DA,DOPAC,HVA,DA/HVA,DA/DOPAC levels .
