摘要
目的:观察淫羊藿苷脂质体经鼻给药能否有效减轻帕金森病模型大鼠黑质区多巴胺能神经元损伤。方法:随机分为假手术组、模型组(PD/in组)、空白脂质体经鼻组(空白PGL/in组)、淫羊藿苷脂质体经鼻组(ICAPGL/in组)、淫羊藿苷经鼻组(ICA/in组)、淫羊藿苷脂质体静脉给药组(ICA-PGL/iv组)。持续干预2周后,用苏木素-伊红(HE)染色法考察淫羊藿苷脂质体对鼻黏膜的安全性。通过大鼠行为学检测,免疫组化法检测黑质区相关指标的变化来观察淫羊藿苷脂质体对帕金森病模型大鼠的疗效。结果:淫羊藿苷脂质体对鼻黏膜无明显损伤。淫羊藿苷各治疗组大鼠经阿朴吗啡诱导后旋转次数较PD/in组大鼠显著减少,双侧前肢功能较PD/in组均有提高,黑质区TH阳性表达较PD/in组显著提高,而黑质区Caspase3阳性表达较PD/in组显著减少,治疗组中以ICA-PGL/in组作用最为显著(P<0.01)。结论:淫羊藿苷脂质体经鼻入脑对帕金森病模型大鼠多巴胺神经元有修复作用,且对鼻黏膜的安全性较好。
Objective :Intranasal administration of icariin propylene glycol-liposome was prepared to investigate the neuro-recovery effects of ICA on hemiparkinsonian rats. Methods :Sham SD rats and PD rats were divided into six groups randomly(Sham SD rats,PD/in rats,blank PGL/in rats,ICA-PGL/in rats,ICA/in rats,ICA-PGL/iv rats,n=8/group)and started daily treatment for two weeks. Histological sections of the rat nasal mucosa after nasal administration of PGL were examined by hematoxylin and eosin(HE).The therapeutic effects of ICA-PGL on 6-hydroxydopamine(6-OHDA)induced hemiparkinsonian rats were assessed by quantifying rotational behavior and stepping test,the levels of tyrosine hydroxylase(TH)and Caspase-3 expressed in substantia nigra(SN)region of hemiparkinsonian rats. Results :Icariin propylene glycol-liposome was high security through intranasal administration with no significant damage to the nasal mucosa. Fewer rotations,more adjusting steps,more TH-positive staining and less Caspase-3 positive staining was seen in ICA administration groups(espacially the ICA-PGL/in rats)than in hemiparkinsonian group. Conclusion :With the advantages of PGL and nose-to-brain pathway,icariin play a repair action on the dopaminogenic neuronal lesion in PD rat induced by 6-OHDA,and shows good security of nasal mucosa.
