摘要
目的分析江苏宿迁地区汉族人群IL-6基因-572C/G多态性与冠心病(CHD)的关系。方法采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)技术检测197例CHD患者及202例健康对照者IL-6基因-572C/G多态性,并进行HardyWeinberg平衡检验,选取部分DNA进行测序,以验证其基因型;用AU5400全自动生化分析仪测定其血糖和血脂水平。结果IL-6-572C/G基因频率在CHD组和健康对照组中均具有群体代表性(P〉0.05);健康对照组和CHD组CC、CG、GG基因型分布分别为53.5%、41.1%、5.4%和36.6%、49.7%、13.7%,C,G等位基因频率则分别为74.0%、26.0%和61.4%,38.6%。与健康对照组比较,CHD组IL-6-572C/G的CG+GG基因型频率显著升高(χ^2=11.53,P=0.007);危险因素分析结果显示,IL-6-572G等位基因增加了CHD患病风险(OR=1.79,95%CI:1.32~2.42,P=0.000)。在CHD组中,血糖血脂水平在IL-6-572CC、CG和GG 3种基因型间的差异无统计学意义(P〉0.05)。早发冠心病与健康对照组比较结果显示CG+GG基因型频率显著升高(χ^2=5.52,P=0.019),且G等位基因频率显著升高(χ^2=6.01,P=0.014);但早发冠心病与晚发冠心病比较,所有基因型差异均无统计学意义(P〉0.05)。结论 IL-6-572G等位基因与江苏宿迁地区汉族人群CHD的发病密切相关,与早发CHD易感性亦显著相关。
Objective The goal in this study was to investigate the association of Interleukin-6( IL-6) gene-572 C / G polymorphism with coronary heart disease( CHD) in Han population of Jiangsu Suqian area. Methods The polymorphism of IL-6 gene-572 C / G was detected using polymerase chain reaction-restriction fragment length polymorphism( PCR-RFLP) method in 197 CHD patients and 202 healthy controls. Meanwhile,the partial genotypes were verified by DNA sequencing analysis. Hardy-Weinberg equilibrium test was performed for statistical analysis. Levels of blood sugar and lipid were determined by AU5400 automatic biochemistry analyzer. Results The characteristic of group representation was found in the gene frequency of IL-6-572 C / G in CHD group and control group( P〈0. 05). The genotype distributions of CC,CG and GG were 53. 5%,41. 1%,5. 4% and 36. 6%,49. 7%,13. 7% in control group and CHD group,respectively. The genotype frequencies of C and G alleles were 74. 0%,26. 0% and 61. 4%,38. 6% in control group and CHD group,respectively. Compared with the control group,the genotype frequency of CG + GG in IL-6-572 C / G was remarkedly increased( χ^2= 11. 53,P = 0. 007). The result of risk factor analysis indicated that IL-6-572 G allele conferred to high risk of CHD( OR = 1. 79,95% CI: 1. 32-2. 42,P = 0. 000). In the CHD group,levels of blood sugar and lipid had no significant difference among the three genotypes of IL-6-572 CC,CG and GG( P〉0. 05). In comparison with the healthy control,the genotype frequency of CG + GG was significantly increased in premature coronary heart disease( PCHD) group( χ^2= 5. 52,P = 0. 019). The frequency of allele G was also greatly increased( χ^2= 6. 01,P = 0. 014). All genotypes had no statistical difference between PCHD group and lateonset coronary heart disease( LCHD) group( P〉0. 05).
