摘要
目的 :观察槐耳颗粒对卵巢癌SKOV-3细胞增殖和凋亡的影响,并探讨其可能的作用机制。方法 :取对数生长期的SKOV-3细胞,用不同质量浓度的槐耳颗粒(0、2、4和8 mg/m L)处理48 h后,在倒置相差显微镜下观察SKOV-3细胞形态的变化;CCK-8法检测槐耳颗粒对SKOV-3细胞增殖的影响;FCM法检测槐耳颗粒对SKOV-3细胞凋亡的影响;蛋白质印迹法检测槐耳颗粒对SKOV-3细胞中凋亡相关蛋白Bcl-2、Bax和cleaved-caspase 3蛋白表达的影响。结果 :用不同质量浓度的槐耳颗粒处理SKOV-3细胞48 h后倒置显微镜下观察发现,各组SKOV-3细胞的形态均发生改变。CCK-8检测结果显示,不同质量浓度的槐耳颗粒能显著抑制SKOV-3细胞的增殖(P<0.05),且呈时间和剂量依赖性;槐耳颗粒能诱导SKOV-3细胞发生凋亡,与对照组相比,早期凋亡率明显增加,差异有统计学意义(P<0.05);蛋白质印迹法检测结果显示,槐耳颗粒可下调Bcl-2蛋白的表达水平(P<0.05),而上调Bax和cleaved-caspase3蛋白的表达水平(P值均<0.05)。结论 :槐耳颗粒在体外可抑制人卵巢癌SKOV-3细胞增殖并诱导其凋亡。
Objective: To investigate the effects of Huaier Particles on proliferation and apoptosis of ovarian cancer SKOV-3 cells in vitro, and to explore its possible mechanism. l^tethods: The SKOV-3 cells were treated with different concentrations of Huaier Particles (0, 2, 4 and 8 mg/mL) for 48 h. Then the morphological changes of SKOV-3 cells were observed under an inverted phase contrast microscope. The proliferation and apoptosis of SKOV-3 cells were detected by CCK-8 method and FCM , respectively. The expression levels of apoptosis-related protein Bcl-2, Bax and cleaved-caspase 3 were detected by Western blotting.Results: The apoptosis-related morphological changes were observed in SKOV-3 cells treated with different concentrations of Huaier Particle. The proliferation of SKOV-3 cells was inhibited by Huaier Particles in time- and dose-dependent manner (P 〈 0.05), and the percentage of early apoptosis cells in Huaier Particle treatment group was significantly increased as compared with that in Huaier Particle-untreated group (P 〈 0.05). The expression levels of Bax and cleaved-caspase 3 proteins were significantly up-regulated (both P 〈 0.05), the the expression level of Bcl-2 protein was significantly down-regulated (P 〈 0.05). Conclusion: Huaier Particles can significantly suppress the proliferation and induce the apoptosis of human ovarian cancer SKOV-3 cells in vitro.
