萱藻中岩藻聚糖硫酸脂的提取工艺优化及其保肝护肝作用 被引量:8

摘要 岩藻聚糖硫酸酯是一种广泛存在于褐藻中的水溶性杂多糖,具有多种生物活性。为研究萱藻岩藻聚糖硫酸脂的保肝护肝作用,以大连沿海的新鲜萱藻Scytosiphon lomentaria为原料,利用复合酶解法提取萱藻中的岩藻聚糖硫酸酯,并用正交试验法优化提取工艺。结果表明:复合酶解法的最佳工艺参数为酶加量2.95%、酶解温度50℃、p H值5.5、酶解时间60 min,在此条件下岩藻聚糖硫酸酯的提取率为4.28%;萱藻岩藻聚糖硫酸酯对CCl4诱导的小鼠急性肝损伤的保肝试验显示,萱藻岩藻聚糖硫酸酯可显著抑制小鼠血清中AST、ALT、LDH活性和肝脏MDA含量的升高,显著提高肝脏GSH含量和SOD活性。研究表明,萱藻中岩藻聚糖硫酸脂具有显著的保肝护肝作用。 Fucoidan as a soluble heteropolysaecharide widely found in brown seaweed was extracted and optimized from fresh seaweed Scytosiphon lomentaria collected in Dalian coast in an orthogonal experiment by an enzymatic method. The results showed that the optimal extraction with crude fucoidan yield of 4.28% was conducted under the condition of the ratio of sample to water = 1:18, the addition of combined enzyme at a rate of 2.95% , hydro-lyzing temperature of 50 ~C , pH value of 5.5, and reaction for 60 rain. The protective effect of the fucoidan on a- cute liver injury induced by CC14 was studied in mice. It was found that the fucoidan from the seaweed had signifi- cant inhibition of the activities of glutamic-oxalacetic transaminase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) and level of malondialdehyde (MDA). The reduced glutathione (GSH) content and superoxide dismutase (SOD) activity were found to be significantly increased, indicating that fucoidan from Scytosi- phon lomentaria showed potential hepatoprotective effect.
