摘要
目的建立HPLC-ELSD法测定香菊胶囊中黄芪甲苷的含量。方法色谱柱:SUPELCO Discovery C18(25 cm×4.6 mm,5μm);流动相:乙腈-水(32∶68);流速:1.0 mL·min-1;柱温:30℃;ELSD参数:漂移管温度为45℃;氮气吹扫流速为490 kPa。采用外标二点法对数方程计算含量。结果黄芪甲苷在0.52~10.40μg进样量对数值与峰面积对数值呈良好的线性关系,回归方程:logY=1.622 logX+4.503,r=0.999 4(n=5);低、中、高3个浓度平均回收率均〉95%,RSD均〈1.6%(n=3)。结论本方法的重复性好、稳定性强、可靠准确,为控制香菊胶囊的质量提供基础。
Objective To establish a method for the content determination of astragaloside Ⅳ in Xiangju capsules by HPLC-ELSD. Methods HPLC-ELSD was adopted. The separation was performed on SUPELCO Discovery C18(25 cm×4.6 mm, 5 μm) column with the mobile phase of acetonitrile-water(32 ∶ 68) at 1.0 mL·min- 1. The column temperature was kept at 30℃. The drift tube temperature of ELSD was 45℃ and the nitrogen fl ow-rate was 490 kPa. Two-point external standard method of logarithmic equation was used to calculate the content of astragaloside Ⅳ. Results A good linearity was showed between the injection volume and peak area at 0.52- 10.4 μg. The regression equation was logY = 1.622 logX + 4.503(r = 0.999 4); the average recoveries were more than 95%(n = 3), and the RSDs were less than 1.6%. Conclusion This method provides the basis for the quality control of Xiangju capsules, with good reproducibility, stability, accuracy and reliability.
