摘要
目的:建立检测康艾注射液中黄芪甲苷含量的HPLC-ELSD法。方法:采用薄层色谱法鉴定黄芪甲苷,采用高效液相色谱法,以蒸发光散射检测器检测,采用外标二点法对数方程计算其含量。色谱柱:SHIMADZUVP-ODS(4.6mm×250mm);流动相:乙腈—水(35:65);流速:1.0mL/min;柱温:30℃;ELSD参数:雾化器温度55℃,漂移管温度为90℃,氮气气压0.75MPa,气体流速为1.15L/min。结果:在色谱图中黄芪甲苷与其它色谱峰完全分离。黄芪甲苷在进样量为0.64~3.20μg时有良好的线性关系Y=1.412X+9.0692(r=0.9999),平均回收率为95.00%,RSD为0.91%。结论:HPLC-ELSD法具有准确、简便、无干扰而且重现性好的优点,适合用于康艾注射液中黄芪甲苷含量的测定。
Objective: To establish a method for determination Astrogloside IV in Kang Ai Injection.Methods: Astrogloside IV was identified by TLC;To determine the quantity by ELSD-HPLC and calculate contents with extenrla standard two-point method and logarithmic equation.Shimadzu VP-ODS column,a mobile phase of acetonitrile-water(35: 65).The flow rate was 1.0mL / min.Room temperature of drift tube for ELSD was 90℃ and the flow rate of nebulizing gas at 1.15L / min.Results: Astrogloside IV chromatographic peak separaed completely.The linear range of Astragaloside IV was 0.64~3.2ug(r=0.9999),The average recoveries was 98.80% and RSD=0.91%.Conclusion: The methods is effective for the quality control of Kang Ai Injection.
