碱性成纤维细胞生长因子及其受体在胎儿和成人皮肤内的表达特征及其意义 被引量:5

摘要 目的 :研究碱性成纤维细胞生长因子 (b FGF)和其受体 (b FGFR)以及磷酸化酪氨酸蛋白 (P Tyr)在胎儿和成人皮肤中的表达特征及其可能的生物学意义。方法 :16例被测标本中包括不同胎龄 (妊娠 12~ 31周 )的胎儿皮肤组织 8例和成人皮肤组织 8例 ,用免疫组织化学方法和病理技术确定 b FGF、b FGFR和 P Tyr这3种蛋白在胎儿和成人皮肤组织中的定位和表达量的变化规律。结果 :b FGF、b FGFR和 P Tyr在胎儿和成人皮肤内都有表达 ,b FGF主要分布于表皮细胞、成纤维细胞、毛囊上皮细胞和血管内皮细胞的胞浆和胞外基质中 ;b FGFR定位于这些细胞的细胞膜上 ;P Tyr蛋白的阳性信号在表皮细胞、成纤维细胞和血管内皮细胞的质膜上和胞浆内都有分布。在胎儿发育初期 ,分别含有这 3种蛋白的阳性细胞数量很低 ;随着胎儿生长发育 ,这3种蛋白的阳性细胞率逐渐增大 ;在发育后期的胎儿和成人皮肤组织中 ,这 3种蛋白表达升高更加显著。结论 :b FGF、b FGFR和 P Tyr在不同发育阶段人的皮肤组织内呈阳性表达 ,显示它们可能对皮肤的发生、结构功能的维持以及伤后修复十分重要。在发育早期的胎儿皮肤中 ,这 Objective:To investigate the localization and expression of basic fibroblast growth factor(bFGF),its receptor(bFGFR) and phosphotyrosine proteins(PTyr) in fetal and adult skins and to explore their potential biological significance.Methods:Skins of 8 fetuses with different gestational ages(1231 weeks) and 8 adults were taken,embedded in paraffin and sectioned.Immunohistochemistry and pathological technics were used to detect the expression intensity and distribution of bFGF,bFGFR,and PTyr.Results:Positive immunohistochemical signals of bFGF,bFGFR,and PTyr could be found in fetal and adult skins,bFGF was mainly located in the cytoplasm and extracellular matrix of epidermal cells,fibroblasts,hair follicle epithelial cells and vascular endothelial cells,EGFR mostly in the cellular membrane of those cells,while PTyr was chiefly located in the cellular membrane and cytoplasm of epidermal cells,fibroblast and endothelial cells.Along with the increase in gestational age,the positive cellular rates of bFGF,bFGFR and PTyr in skin were elevated.In fetus of later stage and adult skins,the positive rates of those three proteins were significantly raised in comparison with younger fetus.Conclusions:The endogenous bFGF and bFGFR might be involved in the cutaneous development in embryonic stage and in the cutaneous structure and function maintenance,and wound healing process in adult stage.The relative lack in protein contents of bFGF,bFGFR and PTyr in skins of younger fetuses might be one of the reasons why the wounds in fetal skins heal by regeneration rather than by scarring.
