摘要
采用斑点杂交技术观察了大鼠烧伤创面细胞内源性重组转化生长因子(transforminggrowthfactorbeta,TGFβ)和其受体基因的转录水平。结果表明,正常皮肤组织存在较弱的上述基因表达。热损伤能诱导TGFβ和TGFβ受体基因表达,它们的表达随着创面修复的进行而逐渐增强,并在创面修复最为活跃的时相达峰值。尔后随着修复的日趋完成与减弱,表达的水平也逐渐下降,但TGFβ受体基因表达的水平先于其配基恢复到或接近正常皮肤的水平。创面细胞TGFβ与其受体基因的共表达表明受体也是创伤愈合过程中的一调节因素。两者在表达模式上的差异提示机体存在生理性的调节机制以限制内源性TGFβ在愈合后期对修复细胞的过度作用。
In the present study,expression of endogenous TGFβ and type Ⅰ,type Ⅱ receptor mRNAs in wound healing of burn rats was observed by dot blot hybridization.Both TGFβ and its type Ⅰ,type Ⅱ receptor mRNAs were found to be expreesive on healing wounds,and their expression appeared to be well regulated.Thermal injury can induce gene expression of TGFβ and its receptors.The expression levels were highest at the time of active repair,which suggests that TGFβ play an important role on epidermis regeneration,that expression of receptors is one of the factors regulating wound healing.The discrepancy between TGFβ and its type Ⅰtype Ⅱ receptor expression at later stages of wound healing implies the presence of well-controled mechanism to limit excessive effect of endogenous TGFβ on repair cells.
